消积抑癌外敷方对Lewis肺癌荷瘤小鼠肿瘤组织TNF-α、MMP-9的影响

摘　　要：目的:分析消积抑癌外敷方对Lewis肺癌荷瘤小鼠肿瘤组织肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-9(MMP-9)的影响。方法:选取健康雄性小鼠30只,采用简单随机分组法以随机数字表法选取10只小鼠设为正常对照组,剩余20只建立Lewis肺癌小鼠模型,随机分为荷瘤对照组、消积抑癌外敷方组,每组10只。建模后8d开始予以干预,荷瘤对照组在瘤体表面外敷生理盐水调和的面糊,消积抑癌外敷方组在瘤体表面外敷消积抑癌外敷方。在治疗30d后,观察小鼠一般状态,并脱颈处死,剥离皮下瘤块,取肺组织,计算各组瘤体重量、瘤重抑制率及肺转移灶数、肺转移抑制率;并比较各组肿瘤组织TNF-α、MMP-9表达水平。结果:治疗30d后,经进食量及5min活动、站立次数评估小鼠一般状态,发现正常对照组平均进食量及5min活动、站立次数均显著高于造模的2组(P<0.05),且消积抑癌外敷方组平均进食量及5min活动、站立次数均高于荷瘤对照组(P<0.05)。治疗30d后,荷瘤对照组瘤体重量及肺转移灶数显著高于消积抑癌外敷方组(P<0.05),消积抑癌外敷方组瘤重抑制率及肺转移抑制率分别为26.63%、53.03%;治疗30d后,荷瘤对照组肿瘤组织TNF-α表达水平显著低于消积抑癌外敷方组(P<0.05),其MMP-9表达水平显著高于消积抑癌外敷方组(P<0.05)。结论:消积抑癌外敷方具有一定抑癌作用,该抑癌作用可能与调控TNF-α、MMP-9表达相关。Objective:To analyze the effect of Xiaoji Yiai Prescription for external application on tumor necrosis factor-α(TNF-α)and matrix metalloproteinase-9(MMP-9)in tumor tissues for tumor-bearing mice with Lewis lung cancer.Methods:30 healthy male mice were selected.Among them,10 mice were selected as a normal controlled group by using simple random grouping of the random number table method,and the remaining 20 mice were established as Lewis lung cancer mice models and were randomly divided into a tumor-bearing controlled group and a Xiaoji Yiai external application group,with 10 mice in each group.The mice started to be given intervention at 8 d after modeling,and the tumor-bearing controlled group was externally applied with a batter mixed with physiological saline on the surface of the tumor,and the Xiaoji Yiai external application group was externally applied with Xiaoji Yiai prescription on the surface of the tumor.After 30 d of treatment,the general status were observed among the mice,and the mice were sacrificed by taking off neck.The subcutaneous masses were stripped off and the lung tissues were taken.The tumor body weight,tumor weight inhibition rate,number of lung metastases and lung metastasis inhibition rate were calculated in each group.The expression levels of TNF-αand MMP-9 in tumor tissues were compared among the groups.Results:After 30 d of treatment,the general status of the mice were evaluated by food intake,5 min activity and standing frequency,and it was found that the average food intake,5 min activity,and standing frequency in the normal controlled group were significantly higher than those in two modeled groups(P<0.05),and the average food intake,5 min activity and standing frequency in the Xiaoji Yiai external application group were higher than those in the tumor-bearing controlled group(P<0.05).After 30 d of treatment,the tumor body weight and the number of lung metastases in the tumor-bearing controlled group were significantly higher than those in the Xiaoji Yiai external application gr
