摘 要:目的:探讨龙葵碱对人乳腺癌MCF-7细胞顺铂敏感性的影响及作用机制。方法:取MCF-7细胞以0、3、6、9、12、15nmol/ml龙葵碱培养48h,采用MTT法检测增殖活性,流式细胞仪检测凋亡情况。取MCF-7细胞分别以0、3、6nmol/ml龙葵碱加上每个浓度使用0、3、6、9、12、15μg/ml梯度浓度的顺铂处理,采用MTT法检测增殖活性,计算不同浓度龙葵碱的顺铂半数抑制浓度(LC50)。Western blot法检测0、3、6nmol/ml龙葵碱加上6μg/ml顺铂培养后化疗敏感相关蛋白ERCC1、PTEN、局部粘着斑激酶(FAK)以及PI3K、Akt、p-Akt蛋白相对表达量,以常规培养MCF-7细胞作为对照组。结果:培养48h后,12、15nmol/ml龙葵碱组MCF-7细胞存活率分别为(86.52±5.46)%和(61.28±7.26)%,与0nmol/ml龙葵碱组相比显著降低(P<0.05);12、15nmol/ml龙葵碱组MCF-7细胞凋亡率分别为(12.68±3.09)%和(18.37±3.52)%,与0nmol/ml龙葵碱组相比显著上升(P<0.05);0、3、6nmol/ml龙葵碱浓度下,顺铂对MCF-7细胞LC50分别为8.65、6.07、4.82μg/ml,浓度依次降低;Western blot结果显示,3、6nmol/ml龙葵碱+顺铂组FAK、PI3K、p-Akt蛋白水平显著低于对照组和0nmol/ml龙葵碱+顺铂组,且6nmol/ml龙葵碱+顺铂组FAK、PI3K、p-Akt蛋白水平更低(P<0.05)。结论:龙葵碱提高人乳腺癌MCF-7细胞对顺铂的敏感性,其作用机制可能与抑制FAK-PI3K/Akt信号通路有关。Objective:To explore the effect and action mechanism of Solanine on the sensitivity of human breast cancer MCF-7 cells to Cisplatin.Methods:MCF-7 cells were cultured with solanine of 0,3,6,9,12 and 15 nmol/ml for 48 h.The proliferation activity was detected by MTT.The apoptosis was detected by flow cytometry.MCF-7 cells were treated with solanine(0,3,6 nmol/ml)and Cisplatin(0,3,6,9,12,15μg/ml).The proliferation activity was detected by MTT.The median inhibition concentration(LC50)of Solanine to Cisplatin was calculated.The relative expression levels of chemotherapy-sensitivity related proteins[ERCC1,PTEN,focal adhesion kinase(FAK)],PI3 K,Akt and p-Akt proteins after culture with solanine(0,3,6 nmol/ml)and 6μg/ml Cisplatin were detected by Western blot.The routine cultured MCF-7 cells were enrolled as a controlled group.Results:After 48 h of culture,survival rates of MCF-7 cells in the 12 and 15 nmol/ml solanine groups were(86.52±5.46)%and(61.28±7.26)%,significantly lower than that in the 0 nmol/ml solanine group(P<0.05).The apoptosis rates of MCF-7 cells in the 12 and 15 nmol/ml solanine groups were(12.68±3.09)%and(18.37±3.52)%,significantly higher than that in the 0 nmol/mL solanine group(P<0.05).Under the conditions of 0,3 and 6 nmol/ml solanine,LC50 values of cisplatin to MCF-7 cells were 8.65,6.07 and 4.82μg/ml,respectively,and the concentration was decreased sequentially.Western blot results showed that proteins levels of FAK,PI3 K and p-Akt in the 3,6 nmol/ml solanine+the Cisplatin group were significantly lower than those in the control group and the 0 nmol/mL solanine+the Cisplatin group,which were lower in the 6 nmol/ml solanine+the Cisplatin group than those in the 3 nmol/ml solanine+the Cisplatin group(P<0.05).Conclusion:Solanine can improve the sensitivity of human breast cancer MCF-7 cells to Cisplatin,and the action mechanism may be related to inhibiting FAK-PI3 K/Akt signaling pathway.
