基于健脾活血法探讨参菝抗瘤液对人胃癌SGC-7901细胞增殖及凋亡的影响

摘　　要：目的:研究参菝抗瘤液抑制人胃癌SGC-7901细胞增殖和诱导凋亡的作用。方法:依据健脾活血法创立的参菝抗瘤液作用于人胃癌SGC-7901细胞,运用MTT法观察药液对SGC-7901细胞增殖的影响,流式细胞仪检测药液对SGC-7901细胞凋亡的影响。结果:(1)MTT法:参菝抗瘤液对人胃癌SGC-7901细胞增殖有一定的抑制作用,且这种抑制作用具有量-效关系,剂量越大,抑制作用越强;(2)流式细胞仪检测:SGC-7901细胞经过参菝抗瘤液作用72h后,与对照组比较,均出现不同程度的凋亡,且总凋亡率随着剂量的增加而增加。结论:参菝抗瘤液通过抑制人胃癌SGC-7901细胞增殖及诱导凋亡发挥其抗癌效应,且与剂量正相关。Objective:To investigate the inhibition of cell proliferation and induction of apoptosis effects of Shenba Antitumor Decoction in human gastric carcinoma cell SGC-7901.Methods:Shenba Antitumor Decoction was founded based on invigorating the spleen and promoting blood flow methods.The MTT assay was used to detect the growth inhibition rates of SGC-7901 cells at 72 h which cultured with Shenba Antitumor Decoction in different concentrations,and flow cytometry was used to observe the cell apoptosis rate.Results:(1)Shenba Antitumor Decoction inhibited the proliferation of human gastric cancer cell line SGC-7901.The different concentrations of Shenba Antitumor Decoction had cytotoxicity onthe SGC-7901 cells,which were dose-dependent.(2)Cells treated by Shenba Antitumor Decoction induced apoptosis.The apoptosis rate were also dose-dependent.Conclusion:Shenba Antitumor Decoction can obviously inhibit SGC-7901 cellular proliferation and induce apoptosis,which are correlated with the concentration.
