摘  要:为优化桑树外植体组织培养体系,以添加质量浓度0~4.0 mg/L的6-BA为条件,调查印度桑品种K2(Morus indica cv. K2)在B5、DKW、MS和WPM培养基中外植体继代培养的增殖形态、生理指标与外植体内6-BA浓度的相关性。结果表明,外植体在WPM培养基添加0.2 mg/L IBA和2.0 mg/L 6-BA时增殖效果最好,30 d时质量增殖倍数最大值为9.32,芽净增殖数最大值为24.67;可溶性蛋白质量分数为3.146%(m_(f)),叶绿素质量分数为0.359 mg/g(m_(f));外植体内6-BA质量浓度最大值达到46.053 ng/L,显著高于其他培养基中外植体内的浓度。由此表明,WPM培养基促进桑树外植体对外源6-BA的吸收,进而使外植体内可溶性蛋白和叶绿素维持相对较高水平,促进了生长发育。本研究为探索桑树组织培养的生长调控机制提供了一种新的研究思路。In order to optimize the tissue culture system of mulberry explants, with the condition of adding 6-BA at a mass concentration of 0-4.0 mg/L, we investigated the correlation between the proliferation morphology, physiological indicators and the 6-BA concentration of the explants of the Indian mulberry cultivar K2(Morus indica cv.K2) in B5, DKW, MS and WPM culture media. The results showed that the proliferation effect of explants performed the best when 0.2 mg/L IBA and 2.0 mg/L 6-BA were added to WPM medium. The maximum proliferation times for 30 d was 9.32 and the maximum bud net multiplication number was 24.67;soluble protein content was 3.146%(m;), and the chlorophyll content was 0.359 mg/g(m;). The maximum concentration of 6-BA in the explants reached 46.053 ng/L, which was significantly higher than the concentration of explants in other media. This shows that WPM promotes the absorption of exogenous 6-BA in mulberry explants, thereby maintaining a relatively high level of soluble protein and chlorophyll in the explants, and promoting growth and development. This study provides a new research idea for exploring the growth regulation mechanism of mulberry tissue culture.
