摘　　要：目的:建立超高效液相色谱串联三重四级杆质谱(UPLC-MS/MS)法同时测定狗皮膏中大黄素、芦荟大黄素2种大黄蒽醌类有效成分的含量。方法:样品经甲醇溶液超声提取后,采用ACQUITY BEH C18柱(100 mm×2.1 mm,1.7μm)进行检测,流动相组成:乙腈(A)-0.1%乙酸(B),梯度洗脱(0~3 min,30%~60%A,3~10 min,60%~60%A),流速为0.3 m L·min^(-1),柱温30℃。质谱采用电喷雾电离源,多重反应检测模式(MRM)扫描。结果:该方法在选定浓度范围内线性关系良好r>0.9996,平均回收率为98.85%~98.91%,精密度RSD≤3.68%(n=6)。结论:该方法可以用于狗皮膏中两种大黄蒽醌类成分的含量测定,为后续的分析提供参考。Objective:To establish a UPLC-MS/MS method for the simultaneous determination of emodin and aloe emodin in Goupi Plaster by UPLC-MS/MS.Methods:The samples were extracted by ultrasonic with methanol solution,ACQUITY BEH C18 column(100 mm×2.1 mm,1.7μm)was used for the detection.The mobile phase consisted of 0.1%acetic acid-acetonitrile and gradient elution was at a flow rate of 0.3 m L·min^(-1).The column temperature was at 30℃.ESI-MS was applied and MRM was used for the scan.Results:The contents of two anthraquinones in Goupi Plaster were determined.The linear relationship was good in the selected concentration range(r>0.9996).The average recovery was 98.85%~98.91%,RSD≤3.68%(n=6).Conclusion:The method can be used for the determination of anthraquinones in Goupi Plaster,and it provides reference for subsequent analysis.
