摘 要:目的研究红毛七的皂苷类化学成分。方法采用硅胶柱色谱、反相ODS柱色谱、Sephadex LH-20凝胶柱色谱和半制备高效液相色谱等方法,从红毛七70%乙醇提取物的正丁醇萃取部位分离化合物,通过波谱分析和化学手段鉴定化学结构。结果分离得到5个三萜皂苷类成分,分别鉴定为3-O-β-D-吡喃葡萄糖-(1→2)-α-L-吡喃阿拉伯糖-白桦酸-28-O-α-L-吡喃鼠李糖-(1→4)-β-D-吡喃葡萄糖-(1→6)-β-D-吡喃葡萄糖苷(1)、HN-saponin F(2)、红毛七皂苷D(3)、3-O-β-D-吡喃葡萄糖-(1→2)-α-L-吡喃阿拉伯糖-齐墩果酸-28-O-α-L-吡喃鼠李糖-(1→4)-β-D-吡喃葡萄糖-(1→6)-β-D-吡喃葡萄糖苷(4)和arboreaside D(5)。结论化合物1为新化合物,且首次在红毛七属植物中发现羽扇豆烷型三萜皂苷,化合物5为首次从该属植物中分离得到。Objective To determine the saponin constituents from the roots and rhizomes of Caulophyllum robustum.Methods The compounds from the n-butanol fraction after 70%ethanol extraction were isolated and purified by column chromatographies of silica gel,ODS,Sephadex LH-20 and semi-preparative HPLC.Their structures were identified by spectroscopic analysis and chemical methods.Results Five triterpenoid saponins were isolated and identified as 3-O-α-L-glucopyranosyl-(1→2)-α-L-arabinopyranosyl-betulinic acid-28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl ester(1),HN-saponin F(2),auloside D(3),3-O-β-D-glucopyranosyl-(1→2)-α-L-arabinopyranosyl-oleanolic acid-28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl ester(4),and arboreaside D(5),respectively.Conclusion Compound 1 is a new compound and this lupane-type triterpenoid saponin has been firstly found from genus Caulophyllum.Compound 5 was obtained from the genus for the first time.
