摘 要:目的建立更贴近生理状态的肝模型对中药注射液进行肝毒性评估。方法建立了一个通过3D打印和微流控芯片组合利用人源细胞的类肝脏系统,对3种市售中药注射液进行肝毒性评价。结果该微流控芯片系统中的内皮细胞可长期培养并模拟血管内皮剪切力。生物打印的3D肝微球经14 d培养后仍能保持正常的白蛋白(ALB)和细胞色素P4503A4酶(CYP3A4)的表达。中药注射剂的肝毒性评价结果表明基于3D打印的类肝脏芯片比2D细胞模型更准确。结论本法准确可信,可为中药肝毒性检测提供新方法。Objective To establish a liver model close to the physiological state to evaluate the hepatotoxicity of Chinese medicine injection.Methods A liver-like system that utilized human cells through a combination of 3D printing and microfluidic chips was built to evaluate the hepatotoxicity of 3 Chinese medicine injections.Results The endothelial cells in the microfluidi chip system could be cultured for a long time and the endothelial shear force simulated.The bioprinted 3D liver microsphere maintained normal expression of albumin and cytochromeP4503A4 enzyme after 14 days of culture.The liver-like chip based on 3D printing was more accurate than the 2D cell model.Conclusion This method is accurate,and can be used to determine the hepatotoxicity of Chinese medicine.
