摘　　要：目的解析头痛宁胶囊在大鼠体内的分布特征。方法采用UHPLC-Q Exactive Focus MS/MS分析大鼠灌胃头痛宁胶囊后不同时间点血浆、不同组织及排泄物中的原形成分分布特征。结果从给药后15、30、60、90、120 min的大鼠血浆样品中分别检出39、44、42、41、38种头痛宁胶囊的原形成分,由此可见,给药30 min后的入血原形成分最多;从心、肝、脾、肺、肾、脑、粪便、尿液中分别检测到7、9、20、4、11、8、21、21种原形成分。这些成分类型主要为黄酮、有机酸、酚及二苯乙烯苷类化合物,包括升麻素、升麻素苷、白藜芦醇、溶血磷脂酰胆碱、天麻素、5-O-甲基维斯阿米醇苷、落新妇苷和2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷等成分。结论本法为后续头痛宁胶囊的药效物质基础研究及药效标志物的筛选提供了重要依据。Objective To analyze the distribution characteristics of Toutongning capsules in rats.Methods UHPLC-Q Exactive Focus MS/MS was used to analyze the distribution characteristics of prototype components in the plasma,tissues and excreta at different time points in rats after the intragastric administration of Toutongning capsules.Results Totally 39,44,42,41 and 38 prototype components of Toutongning capsules were detected in the plasma samples of rats at 15,30,60,90 and 120 min after the administration,respectively.It can be seen that prototype components of Toutongning capsule were the most 30 min after the administration.Totally 7,9,20,4,11,8,21 and 21 prototype components were detected in the heart,liver,spleen,lung,kidney,brain,excrement and urine,respectively.These compounds were mainly flavonoids,organic acids,phenols and stilbene glycosides,including cimifugin,cimicifugoside,resveratrol,lysophosphatidyl choline,gastrodin,5-O-methylvisammil glycoside,astilbin and 2,3,5,4’-tetrahydroxy stilbene-2-O-β-D-glucoside.Conclusion This method provides an important basis for the basic research of pharmacodynamic substances and the screening of pharmacodynamic markers of Toutongning capsules in the future.
