摘 要:目的:鉴定不同产区胎菊、朵菊的主要差异化学成分,为杭白菊的道地性研究提供依据。方法:收集不同产区的杭白菊胎菊和朵菊样品,采用超高效液相色谱-四级杆-飞行时间质谱法(UPLC-Q-TOF-MS)进行化学成分分析,综合运用数据非依赖扫描(DIA)与数据依赖扫描(DDA),结合本地及在线数据库,鉴定主要化学成分。应用正交偏最小二乘法-判别分析(OPLS-DA)等多元统计分析方法,对胎菊与朵菊、不同产区胎菊与朵菊、道地产区(桐乡)与非道地产区胎菊与朵菊进行模式分类,以分析过程中变量重要性投影(VIP)值、t检验筛选出主要的差异化合物。结果:从供试样品中共鉴定出33个化合物。胎菊与朵菊化学轮廓差异明显,D-甘露糖、羟基芫花素、槲皮素等为主要的差异化合物;不同产区胎菊化学轮廓差异较明显,其中桐乡胎菊中鸟苷、姜糖酯B等成分含量显著高于其他产区,可作为鉴定道地产区的标志性成分;不同产区朵菊的差异相对较小,但浙江和其他省份(江苏、河南)产朵菊有一定差别,红车轴草素-7-O-β-D-葡萄糖苷为主要的差异化合物。结论:不同产区胎菊的差异较大,而朵菊的差异较小,可能的原因为杭白菊成熟后,化学成分的含量趋于稳定。Objective:This paper identified the differential chemical components in dried buds(Taiju)and dried capitulum(Duoju)of Chrysanthemum morifolium cv.'Hangju'from different producing areas,which is expected to lay a basis for studying the geoherbalism of Chrysanthemi Flos derived from Ch.morifolium cv.'Hangju'(hereinafter referred to as Hangju).Metheds:Ultra performance liquid chromatography-quadrupole-time of flight-mass spectrometry(UPLC-Q-TOF-MS),coupled with data-independent acquisition(DIA),data-dependent acquisition(DDA),and local and online databases,was employed for chemical component identification of Taiju and Duoju from different producing areas.Orthogonal partial least squares discriminant analysis(OPLS-DA)was used for the classification of Taiju and Duoju,Taiju/Duoju from different producing areas,and Taiju/Duoju from genuine producing area(Tongxiang)and non-genuine producing areas.The differential markers were screened based on VIP value and t-test.Results:A total of 33 compounds were identified from the samples.The chemical components were obviously different between Taiju and Duoju,and D-mannose,hydroxygenkwanin,and quercetin were the major differential components.The chemical component profile of Taiju was significantly different among the producing areas.To be specific,content of guanosine and ginger glycolipid B in Taiju from Tongxiang was significantly higher than that from other areas,and thus the two can be used as makers for the geoherbalism of Taiju.Chemical components of Duoju from different areas showed little difference.However,Duoju from Zhejiang was different from that of other areas(Jiangsu,Henan),with the differential compound of pratensein-7-O-β-D glucoside.Conclusion:There are greater differences in Taiju from different producing areas,but smaller differences in Duoju,which may be indebted to the stable content of chemical components in the mature capitulum.
