摘  要:羊传染性脓疱病毒(orf virus,ORFV)可引起动物口唇等黏膜上皮异常角化,导致严重的增生性病变。由于临床存在复杂的患病风险,使ORFV感染早期对症治疗至关重要。因此,研究病毒对细胞增殖与分化的影响有助于筛选抗病毒路径。研究表明,角质形成细胞的增殖和分化进程直接受到角蛋白(CKs)等分子转换调控。为研究ORFV感染对CKs转换的影响,试验室利用ORFV-HaCaT细胞体外感染模型,通过RT-PCR方法检测病毒感染的不同时间段DSG3、Ki67和CKs转录水平。结果显示,DSG3、Ki67、CK2分子表达一直处于稳定,而CK1/CK10、CK5/CK14则在ORFV感染早期相继发生短暂的正向波动,但很快又受到病毒抑制,表明病毒会持续调节感染诱导影响细胞分化趋势。研究为深入探讨ORFV引起角质细胞CKs波动的分子机制奠定了基础。Orf virus(ORFV)can cause abnormal keratosis of the mucosal epithelium where on the mouth and lip of animals,leading to severe hyperplastic lesions.Because of the complex clinical risk,early symptomatic treatment of ORFV infection was very important.Therefore,studying the effect of virus on cell proliferation and differentiation was helpful to screen antiviral pathways.Studies had shown that the proliferation and differentiation processes of keratinocytes were directly regulated by molecular transformations such as keratin(CKs).To investigate the effect of ORFV infection on CKs conversion,ORFV-HaCaT cell infection model in vitro was used to detect the transcription levels of DSG3、Ki67 and CKS at different time periods of viral infection by RT-PCR.Results showed that DSG3、Ki67、CK2 molecular expression had been in stable,whereas CK1/CK10 and CK5/CK14 occurred in the early infection of ORFV brief positive fluctuation,but quickly by virus inhibition,suggesting that the virus will continue to adjust the infection induced by affecting cell differentiation trend.It provided a basis for further study of the molecular mechanism of keratinocyte CKS fluctuations induced by ORFV.
