摘 要:目的探讨miR-140与Beclin1在不同退变程度的软骨组织内表达的变化并分析两者表达的相关性。方法选取因骨关节炎(OA)行膝关节置换患者20例,依据Kellgren&Lawrence(K&L)分级标准分为两组,各10例。对照组为股骨颈骨折行髋关节置换的正常股骨头软骨10例。HE染色、Masson染色、番红固绿染色评估软骨的形态,RT-PCR检测各组软骨内的miR-140与Beclin1的表达,Westernblot检测软骨miR-140、Beclinl蛋白表达。结果与股骨头正常软骨组织相比,OA组软骨组织的HE染色、Masson染色、番红固绿染色显示关节软骨出现退变退变,且退变程度与K&L分级标准分级相一致。通过RT-PCR、Westernblot检测软骨组织miR-140、Beclin1的表达量,发现OA软骨组织miR-140、Beclin1表达水平明显低于股骨头正常软骨组织(P<0.01);且OA组软骨依据K&L分级,退变程度越高,上述两种基因表达量越低(P<0.01)。统计学分析不同软组织miR-140、Beclin1表达水平,两者的表达具有正相关性。结论miR-140、Beclin1的表达随着软骨退变程度加重而下降,miR-140可能通过影响Beclin1的表达而引起软骨的退变。Objective To investigate the changes of miR-140 and Beclin1 expression in cartilage tissues with different degrees of degeneration and analyze the correlation between the expression of miR-140 and Beclin1.Methods 20 patients with osteoarthritis(OA)who underwent knee replacement in Baiyun Hospital affiliated to Guizhou Medical University were selected.According to Kellgren&Lawrence(K&L)classification standard,they were divided into two groups with 10 patients in each group,and 10 patients in the control group with normal femoral head cartilage who underwent hip replacement for femoral neck fracture.Morphology of cartilage was evaluated by HE staining,Masson staining and pinkish green staining.Expressions of miR-140 and Beclin1 in each group were detected by RT-PCR,and expressions of miR-140 and Beclinl proteins in cartilage were detected by western blot(WB).Results Compared with normal femoral head cartilage tissue,HE staining,Masson staining and Panther-fixation green staining of cartilage tissue in OA group showed degeneration of articular cartilage,and the degree of degeneration was consistent with K&L grading standard.The expression levels of miR-140 and Beclin1 in cartilage tissues were detected by RT-PCR and western blot(WB),and it was found that the expression levels of miR-140 and Beclin1 in OA cartilage tissues were significantly lower than that in normal femoral head cartilage tissues,and the difference was statistically significant(P<0.01).Moreover,cartilage in the OA group was graded according to K&L.The higher the degree of degeneration,the lower the expression levels of the above two genes were,and there were statistical significantly differences(P<0.01).Statistical analysis showed that the expression levels of miR-140 and Beclin1 in different soft tissues were positively correlated.Conclusion the expressions of miR-140 and Beclin1 decreased with the aggravation of cartilage degeneration,and miR-140 may cause cartilage degeneration by affecting Beclin1 expression.
