摘 要:目的探讨微小核糖核酸-519(miRNA-519)抑制人类抗原R(HuR)、血管内皮生长因子(VEGF)和基质金属蛋白酶9(MMP9)表达对缺血下肢血管新生的影响。方法选取72只SD大鼠,按照随机数字表法分为对照组、miRNA-519过表达组和miRNA-519抑制剂组。统计并比较三组SD大鼠下肢血流量激光多普勒血流系数、毛细血管数量计数、毛细血管密度、组织中HuR、VEGF和MMP9蛋白的表达。结果第21天,miRNA-519过表达组的SD大鼠下肢血流激光多普勒血流系数、HuR、VEGF和MMP9蛋白表达最低,毛细血管数量和密度最少,与其他两组比较,差异有统计学意义(P<0.05);miRNA-519抑制剂组的大鼠下肢血流激光多普勒血流系数、HuR、VEGF和MMP9蛋白表达最高,毛细血管数量和密度最多,差异有统计学意义(P<0.05)。结论miRNA-519对血管生成具有重要的作用,其机制可能与抑制HuR、VEGF和MMP9表达相关。Objective To investigate the inhibitory effect of microRNA-519(miRNA-519)on the expression of human antigen R(HuR),vascular endothelial growth factor(VEGF),matrix metalloproteinase 9(MMP9)and its impact on the angiogenesis of ischemic lower limbs.Method A total of 72 SD rats were selected and divided into the control group,miRNA-519 overexpression group,and miRNA-519 inhibitor group according to the random number table method.The lower limb’s laser Doppler blood flow velocity,capillary number count,capillary density,and the expression of HuR,VEGF,and MMP9 protein in the relevant tissues were counted and compared between different groups.Result On day 21,SD rats in the miRNA-519 overexpression group occupied the lowest laser Doppler blood flow velocity of the lower limb,lowest expression levels of HuR,VEGF,and MMP9 protein,and the smallest number and lowest density of capillaries.The differences were statistically significant(P<0.05).In the miRNA-519 inhibitor group,the expression of laser Doppler blood flow velocity,HuR,VEGF,and MMP9 protein of lower limb blood flow reached the highest levels,and both their number and density of capillaries were also the highest,and the differences were statistically significant(P<0.05).Conclusion miRNA-519 plays an essential role in angiogenesis,and its underlying mechanism may be related to its inhibitory effect on HuR,VEGF,and MMP9.
