扶正固本汤联合穴位贴敷对咳嗽变异性哮喘大鼠IL-4、IL-10、TGF-β的影响研究

摘　　要：目的通过观察扶正固本汤联合穴位贴敷对咳嗽变异性哮喘大鼠肺泡灌洗液中白细胞介素-4(IL-4)、白细胞介素-10(IL-10)、转化生长因子-β(TGF-β)水平的影响,阐述其治疗咳嗽变异性哮喘的部分机制。方法将40只大鼠随机分成空白组、模型组、地塞米松组、治疗组,每组10只,采用OVA联合Al(OH)_(3)多次激发致敏法造模。治疗组给予扶正固本汤6g/(kg·d)灌胃,"伏九贴"贴于相应穴位,每次1.5h,每日1次。地塞米松组给予0.5mg/(kg·d)灌胃。空白组、模型组生理盐水灌胃,生理盐水浸纱布贴在相应穴位。疗程14d。观察大鼠体征、咳嗽次数,HE染色观察肺组织病理结构,ELISA法检测肺泡灌洗液中IL-4、IL-10、TGF-β含量。结果1)经治疗后大鼠咳嗽次数减少,与模型组比差异有统计学意义(P<0.05);2)经治疗后地塞米松组、治疗组IL-4水平降低,IL-10、TGF-β水平升高,与模型组比差异有统计学意义(P<0.05);地塞米松组与治疗组之间IL-4、TGF-β含量无明显差异(P>0.05);治疗组IL-10含量高于地塞米松组(P<0.05)。结论扶正固本汤联合穴位贴敷能够通过提高IL-10、TGF-β表达,抑制IL-4的表达,减轻气管炎症。Objective:To observe the effect of Fuzheng Guben Decoction combined with acupoint application on the level of IL-4,IL-10,TGF-β in alveolar lavage fluid of rats with cough variant asthma,and to elaborate the partial mechanism of it in the treatment of cough variant asthma.Methods:A total of 40 rats were randomly divided into the blank group,model group,dexamethasone group,and treatment group,10 rats in each group.OVA combined with Al(OH)_(3) multiple excitation sensitization method was used to establish the model.The treatment group was given Fuzheng Guben Decoction 6 g/(kg·d)by intragastric gavage,and Fujiu Paste was applied to the corresponding acupoints,once a day,1.5 h/time.The dexamethasone group was intragastrically administered 0.5 mg/(kg·d),normal saline-soaked gauze was intragastrically administered at the corresponding acupoints in the blank group,the model group was intragastrically administered 0.9%normal saline,and the normal saline-soaked gauze was intragastrically administered at the corresponding acupoints.The period of treatment was 14 days.The cough times of rats were observed.The pathological structure of lung tissue was observed by HE staining.The content of IL-4,IL-10,TGF-β in alveolar lavage fluid was detected by ELISA method.Results:1)The cough frequency of rats was decreased after internal and external treatment with Chinese medicine,and the difference was statistically significant compared with model group(P<0.05);2)After treatment,IL-4 in the dexamethasone group and in the treatment group was decreased and the level of IL-10,TGF-β was increased,and the difference was statistically significant compared with the model group(P<0.05);the contents of IL-4 and TGF-β in dexamethasone group were not significantly different from those in the treatment group(P>0.05).The level of IL-10 in treatment group was higher than that in dexamethasone group(P<0.05).Conclusion:Fuzheng Guben Decoction combined with acupoint application can improve the expression of IL-10,TGF-β ,inhibit the expression of
