五味消痛酊治疗急性闭合性软组织损伤、抗炎、镇痛作用的实验研究

摘　　要：目的探讨五味消痛酊治疗急性闭合性软组织损伤、抗炎、镇痛作用。方法将32只大鼠随机分为空白对照组、模型对照组、五味消痛酊组、扶他林软膏组共4组,除空白组外将其余各组大鼠固定在软组织打击器上通过打击锤敲打造成大鼠急性闭合性软组织损伤,在造模当天给药;分别在给药的第1、3、5、7天测定大鼠损伤症状指数和疼痛缩足反射阈值;在第7天末次给药后解剖大鼠,腹主动脉取血,测定全血黏度、血浆黏度及血细胞比容,取受伤组织测定PGE-2、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的表达水平。结果与模型对照组相比,给药的第3、4、7天,五味消痛酊均可显著降低大鼠损伤症状指数,显著升高大鼠疼痛缩足反射阈值;与模型对照组相比,给药的第7天,五味消痛酊可显著降低全血低切(10/s)、中切(60/s)、高切(150/s)黏度,血浆黏度和血细胞比容,显著降低损伤组织中PGE-2、TNF-α、IL-1β的表达水平。结论五味消痛酊对于急性闭合性软组织损伤具有一定治疗作用,可以起到镇痛作用并且可以通过降低PGE-2、TNF-α、IL-1β的表达来改善炎症反应。Objective:To investigating the effect of Wuwei Xiaotong Tincture on acute closed soft tissue injury,anti-inflammation and analgesia.Methods:32 rats were randomly divided into 4 groups:blank control group,model control group,Wuwei Xiaotong Tincture group and voltalin ointment group.Except for the blank group,the rats in other groups were fixed on the soft tissue percussion device and made into acute closed soft tissue injury by percussion hammer.The injury symptom index was measured on the 1,3,5 and 7 d after administration.The threshold of paw constriction reflex was measured on the 1,3,5 and 7 d after administration.After the last administration on the 7 th day,the rats were dissected,and the blood was taken from the abdominal aorta.The whole blood viscosity,plasma viscosity and hematocrit of the rats in each group were measured.The expression levels of PGE-2,TNF-α,IL-1β and IL-6 in the injured tissues were measured.Results:Compared with the model control group,Wuwei Xiaotong Tincture could significantly reduce the injury symptom index of rats and significantly increase the pain foot retraction reflex threshold of rats after 3,4 and 7 d of administration.Compared with the model control group,on day 7 of administration,Wuwei Xiaotong Tincture could significantly reduce the whole blood low shear(10/s),middle shear(60/s),high shear(150/s)viscosity,plasma viscosity and hematocrit,and significantly reduce the expression levels of PGE-2,TNF-α and IL-1β in rat tissues.Conclusions:Wuwei Xiaotong Tincture has a certain therapeutic effect on acute closed soft tissue injury,can play an analgesic role,and can improve the inflammatory response by reducing the expression of PGE-2,TNF-αand IL-1β.
