摘　　要：目的探讨白细胞介素-12B(IL-12B)、上皮型钙黏素(E-cadherin)和12(R)-脂加氧酶(ALOX12B)在宫颈上皮内瘤变(CIN)组织中的表达及其与高危型人乳头瘤病毒(HPV)感染的关系。方法选取2019年7月-2020年7浙江省台州医院收治的238例CIN患者为研究组,同期进行宫颈组织活检的188例宫颈炎患者为对照组。研究组患者根据宫颈病变程度,分为Ⅰ级组59例、Ⅱ级组137例、Ⅲ级组42例;根据HPV感染情况,分为无感染组38例、低危HPV感染组49例、高危HPV感染组151例,根据ALOX12B mRNA水平检测情况分为高表达组187例和低表达组51例。采用免疫组化法和聚合酶链反应(PCR)测定两组宫颈组织中IL-12B、E-cadherin和ALOX12B蛋白和表达mRNA含量,分析其与CIN级别、HPV感染的关系。结果研究组ALOX12B免疫组化乘积和mRNA含量均高于对照组,IL-12B和E-cadherin免疫组化乘积和mRNA含量均低于对照组(P<0.05)。Ⅲ级组患者ALOX12B免疫组化乘积,mRNA含量高于Ⅰ级组和Ⅱ级组;Ⅲ级组患者IL-12B和E-cadherin免疫组化乘积、mRNA含量均低于Ⅰ级组(P<0.05)。高危HPV感染组患者ALOX12B免疫组化乘积、mRNA含量均高于无感染组,IL-12B和E-cadherin表达低于无感染组(P<0.05)。ALOX12高表达患者CIN宫颈组织中PI3K、ERK1、Ki67的mRNA含量高于ALOX12B低表达组(P<0.05)。结论 CIN宫颈组织中ALOX12B表达上调、IL-12B和E-cadherin表达下调或缺失,可能参与高危HPV感染导致宫颈癌的发生发展过程。OBJECTIVE To explore the expressions of interleukin-12 B(IL-12 B),epithelial cadherin(E-cadherin) and 12(R)-lipoxygenase(ALOX12 B) in cervical intraepithelial neoplasia(CIN) and analyze their relationship with high-risk human papilloma virus(HPV) infection.METHODS A total of 238 patients with CIN who were treated in Taizhou Hospital from Jul.2019 to Jul.2020 were assigned as the study group,meanwhile,188 patients with cervicitis who received cervical tissue biopsy were chosen as the control group.The patients of the study group were divided into the grade Ⅰ group with 59 cases,the grade Ⅱ group with 137 cases and the grade Ⅲ group with 42 cases according to the severity of cervical lesions.The patients were divided into the no infection group with 38 cases,the low-risk HPV infection group with 49 cases,and the high-risk HPV infection group with 151 cases according to the prevalence of HPV infection,and the patients were divided into the high expression group with 187 cases and the low expression group with 51 cases according to the ALOX12 B mRNA level.The expression levels of IL-12 B,E-cadherin and ALOX12 B proteins and mRNA content in cervical tissues were detected by immunohistochemistry and polymerase chain reaction(PCR),and their relationship with grade of CIN and HPV infection was observed.RESULTS The immunohistochemical product and mRNA content of ALOX12 B of the study group were signigifcantly higher than those of the control group,while the immunohistochemical products and mRNA contents of IL-12 B and E-cadherin of the study group were signigifcantly lower than those of the control group(P<0.05).The immunohistochemical product of ALOX12 B in the grade III group,and mRNA content of ALOX12 B of the grade III group was signigifcantly higher than that of the grade I group and the grade II group.The immunohistochemical products and mRNA contents of IL-12 B and E-cadherin of the grade III group were signigifcantly lower than those of the grade I group(P<0.05).The immunohistochemical product and mRNA level
