摘 要:目的本文采用RP-HPLC-UV法对经典名方金水六君煎中橙皮苷的含量进行测定。方法色谱柱选用十八烷基硅烷键合硅胶柱,以甲醇、HAc和水为流动相,比例为35∶4∶61;UV检测波长为283nm,流速设置为1.0mL·min^(-1),进样量为10μL,柱温设置为30℃。结果橙皮苷含量在2~14μg之间呈良好线性关系,回归方程为:Y=20.474X-0.6305,R2=0.9994,确定金水六君煎物质基准中每克含橙皮苷不得少于5.0mg。结论本文建立的橙皮苷含量测定方法稳定可靠,实验操作简便,为中华传统中医药经典名方的研究与发展奠定基础。OBJECTIVE In order to actively respond to the call of the state and vigorously carry forward the culture of traditional Chinese Medicine,the content of hesperidin in Jinshui Liujun Decoction was determined by RP-HPLC-UV.METHODS Octadecylsilane bonded silica gel column was used,the mobile phase was methanol glacial acetic acid water(35∶4∶61);the detection wavelength was 283nm;the flow rate was 1.0mL·min^(-1).The injection volume was 10μL and the column temperature was set at 30℃.RESULTS When the content of hesperidin was between 2μg and 14μg,the linear relationship was good the regression equation was Y=20.474X-0.6305,R2=0.9994.The content of hesperidin in Jinshui Liujun Decoction should not be less than 5.0mg·g^(-1).CONCLUSION The method established in this paper is stable,reliable and easy to operate,which lays a foundation for the research and development of traditional Chinese medicine.
