摘 要:目的探讨超高效液相色谱(UHPLC)对大黄饮片炮制前后化学成分定性定量分析。方法有机溶液提取后采用UHPLC计划对生大黄、熟大黄和大黄炭进行化学成分定性定量分析,观察大黄饮片炮制前后化学变化规律的验证。结果生大黄中苷类物质、缩合鞣质较多,熟大黄及大黄炭中表达降低,没食子酸类物质增加。生大黄中游离蒽醌类升高,在熟大黄及大黄炭中几乎无表达;炮制前后的大黄饮片中苯丁酮类、二苯乙烯苷类、鞣质类成分出现变化。结论说明药理活性与化学成分变化存在一定的关联性。OBJECTIVE To investigate the qualitative and quantitative analysis of chemical components of rhubarb decoction pieces before and after processing by UHPLC.METHOD After the organic solution was extracted,the UHPLC plan was used to qualitatively and quantitatively analyze the chemical components of raw rhubarb,cooked rhubarb and rhubarb charcoal,and observe the verification of the chemical changes before and after the processing of rhubarb decoction pieces.RESULT There are more glycosides and condensed tannins in raw rhubarb,the expression in cooked rhubarb and rhubarb charcoal decreases,and gallic acid substances increase.The free anthraquinones in raw rhubarb increased,and there was almost no expression in cooked rhubarb and rhubarb charcoal.There were changes in the phenylbutanone,stilbene glycosides and tannins in the rhubarb decoction pieces before and after processing.CONCLUSION These results indicated that there was a certain correlation between pharmacological activity and chemical composition change.
