摘  要:目的:探讨中等强度下坡跑对肥胖小鼠脂肪组织慢性炎症的改善及TRIB3/AKT通路和巨噬细胞极化的调节作用。方法:75只5周龄C57BL/6N雄性小鼠随机分为正常组(CON组)和高脂膳食组(HFD组),干预12周后组内随机分为正常安静组(NC组)、正常运动组(NE组)、高脂安静组(HC组)和高脂运动组(HE组)。运动组进行最大运动强度45%的中等强度下坡跑,持续8周。干预完成后,取双侧附睾脂肪并称取湿重,HE染色进行脂肪病理组织学观察,免疫荧光染色检测巨噬细胞极化相关蛋白表达与定位,Western blot方法检测脂肪组织IL-1β、IL-10、iNOS、Arg-1、TRIB3、p-AKT和AKT蛋白表达,ELISA方法检测脂肪组织中炎症相关蛋白水平。结果:1)与NC组相比,HC组附睾脂肪重量、脂肪细胞面积均显著增加(P<0.01);脂肪间质内可见大量呈花冠样结构浸润的炎症细胞且IL-1β蛋白水平显著增加、IL-10蛋白水平显著下降(P<0.01);CD86和iNOS表达均显著增加、CD206和Arg-1表达则显著下降(P<0.01);TRIB3蛋白表达显著增加、AKT蛋白磷酸化水平显著降低(P<0.01)。2)8周中等强度下坡跑干预后,与对应的安静组相比,HE组附睾脂肪重量、脂肪细胞面积均显著下降(P<0.01);HE组脂肪细胞间质中呈花冠样结构浸润的炎症细胞明显减少;NE组和HE组脂肪组织IL-1β表达均显著降低、IL-10表达则显著增加(P<0.01);NE组和HE组CD86和iNOS表达显著降低、CD206和Arg-1表达则均显著增加(P<0.01);NE组和HE组TRIB3表达均显著降低、HE组AKT磷酸化激活水平显著升高(P<0.01)。结论:高脂膳食导致小鼠脂肪组织巨噬细胞M1型极化增加、M2型极化减少,诱发慢性炎症;8周中等强度下坡跑有效改善肥胖小鼠脂肪组织慢性炎症,其机制可能与下坡跑抑制TRIB3/AKT信号进而诱导巨噬细胞M2型极化、减少M1型极化密切相关。Objectives:to observe the effects of moderate-intensity downhill running on chronic inflammation of adipose tissue in obese mice,and the regulatory effects of TRIB3/AKT signaling and macrophage polarization.Methods:seventy-five C57BL/6 N male mice were randomly divided into a control group and a high-fat diet group.After 12 weeks,the mice were randomly divided into a normal control group(NC group),a normal exercise group(NE group),a high-fat diet group(HC group)and a high-fat diet and exercise group(HE group).In the exercise group,8-week moderate-intensity downhill running was performed.The bilateral epididymal fat were taken from the animal and weighed.HE staining was used to observe the histopathology of the fat.Immunofluorescence staining was used to detect the expression and localization of macrophage polarization-related proteins.The expressions of IL-1β,IL-10,iNOS,Arg-1,TRIB3,AKT,p-AKT were detected by Western blot.The IL-1βand IL-10 proteins in visceral fat were detected by ELISA.Results:Compared with the NC group,the weight of epididymal fat and the area of adipocytes in the HC group significantly increased(P<0.01);the corolla-like areas of inflammatory cell infiltration were observed in the adipose tissue of the HC group;the levels of IL-1β,CD86,iNOS and TRIB3 significantly increased in the adipose tissue of the HC group(P<0.01);the levels of IL-10,CD206,Arg-1 and the phosphorylation of AKT significantly decreased in the adipose tissue of the HC group(P<0.01).After 8 weeks of moderate-intensity downhill running intervention,compared with the corresponding sedentary group,the weight of epididymal fat and the area of adipocytes in the HE group significantly decreased(P<0.01);the corolla-like areas of inflammatory cell infiltration decreased in the adipose tissue of the HE group;the levels of IL-1β,CD86,iNOS and TRIB3 significantly decreased in the adipose tissue of the NE group and HE group(P<0.01);the levels of IL-10,CD206,Arg-1 and the phosphorylation of AKT significantly decreased in the adipose tis
