摘  要:目的采用高效液相色谱连用紫外检测器检测经典名方半夏泻心汤中黄芩苷含量。方法色谱柱以十八烷基硅烷键合硅胶为填充剂,以0.22%磷酸水为流动相A,甲醇为流动相B,比例为53∶47,流速设置为1mL·min^(-1),检测波长为280nm。结果黄芩苷出峰时间为10min左右,峰形良好且无其他物质干扰,黄芩苷含量在0.26~1.56μg之间呈良好线性关系,得回归方程为:Y=60.999X-1.3235,R^(2)=0.9998。最终确定半夏泻心汤物质基准的黄芩苷每剂含量范围309.23~574.28mg。结论该方法有效简便的测定出半夏泻心汤物质基准中黄芩苷的含量,为经典名方半夏泻心汤的进一步研究与发展奠定基础。OBJECTIVE In order to determine the content of Baicalin in Banxia Xiexin Decoction by HPLC with UV detector.METHODS The chromatographic column was composed of octadecylsilane bonded silica gel,0.22%phosphoric acid water as mobile phase A and methanol as mobile phase B with the ratio was 53∶47,the flow rate was set at 1mL·min^(-1),and the detection wavelength was 280nm.RESULTS The peak time of baicalin was about 10 minutes,the peak shape was good and there was no interference from other substances.The content of Baicalin showed a good linear relationship between 0.26 and 1.56μg,and the regression equation was Y=60.999X-1.3235,R^(2)=0.9998.Finally,the content range of Baicalin in Banxia Xiexin Decoction was 309.23~574.28mg.CONCLUSION This method is effective and simple to determine the content of Baicalin in Banxia Xiexin Decoction,which lays a foundation for further research and development of Banxia Xiexin Decoction.
