摘  要:目的:评价维血宁合剂提取液醇沉法和膜分离法的精制工艺。方法:本研究以维血宁合剂提取液为研究对象,以物理化学参数、固形物去除率、指标性成分转移率为指标,评价维血宁合剂提取液醇沉法和膜分离法2种不同的精制工艺,通过考察不同截留分子量、操作压力、提取液浓度对膜分离指标以及膜通量的影响,优选最佳的纯化工艺。结果:膜分离法的物理化学参数优于醇沉法。醇沉液指标性成分儿茶素、表儿茶素、虎杖苷、白藜芦醇、槲皮素的转移率分别为82.34%,83.31%,79.06%,77.51%,80.16%,固形物去除率为64.75%。优选膜分离最佳工艺为膜压力0.5 MPa;提取液浓度0.11 g·mL^(-1);截留分子量大小30 kDa,膜分离液指标性成分儿茶素、表儿茶素、虎杖苷、白藜芦醇、槲皮素的转移率分别为83.44%,84.32%,80.42%,83.36%,83.58%,膜通量为10.57 mL·m^(-2)·min^(-1),固形物去除率为65.96%。结论:相比于醇沉法,膜分离法更有利于维血宁合剂提取液的精制,为维血宁合剂工业化生产奠定基础。Objective:To evaluate the purification process of Weixuening mixture extract by alcohol precipitation method and membrane separation method.Methods:In this study,the Weixuening mixture extract was used as the research object,and physicochemical parameters,solid removal rate,and the transfer rates of indicative components were used as the indexes to evaluate the two different refining processes of Weixuening mixture extract,i.e.,alcohol precipitation method and membrane separation method.By investigating the influences of different molecular weight cut-offs,operating pressure,and extracting solution concentration on membrane separation index and membrane flux,the best purification process was selected.Results:The physicochemical parameters of the membrane separation method were significantly better than those of the alcohol precipitation method.The transfer rates of the indicative components of the alcohol precipitation solution such as catechin,epicatechin,polydatin,resveratrol,and quercetin,respectively,were 82.34%,83.31%,79.06%,77.51%,and 80.16%,respectively.The solid removal rate was 64.75%.The best process for membrane separation was as follows:the membrane pressure was 0.5 MPa;the extract concentration was 0.11 g·mL^(-1);the molecular weight cutoff was 30 kDa.The transfer rates of the indicative components such as catechin,epicatechin,polydatin,resveratrol,and quercetin in the membrane separated solution were 83.44%,84.32%,80.42%,83.36%,and 83.58%,respectively.The membrane flux was 10.57 mL·m^(-1)·min^(-1) and the solid removal rate was 65.96%.Conclusion:Compared with the alcohol precipitation method,the membrane separation method is more conducive to the refining of the extract of Weixuening mixture,which lays the foundation for the industrial production of Weixuening mixture.
