摘 要:目的研究彭县雪胆Hemsleyapengxianensis中的三萜类化学成分。方法采用硅胶柱色谱法、半制备高效液相色谱等方法进行分离纯化,结合NMR、IR、MS等波谱技术对其结构进行鉴定,并运用MTT法测定化合物对He La细胞的细胞毒活性。结果从雪胆中分离得到葫芦烷三萜类化合物17个,结构分别鉴定为2α,3α,20-三羟基葫芦素-16,25-二乙酰氧基-5-烯-7,11,22-三酮(1)、jinfushanencin B(2)、短柄雪胆苷D(3)、scandenogenin D(4)、jinfushanencin C(5)、jinfushanoside D(6)、hexanorcucurbitacin F(7)、短柄雪胆苷A(8)、cucurbitacin V(9)、hemslepenside A(10)、jinfushanencin F(11)、scandenoside R3(12)、scandenoside R2(13)、巨花雪胆G(14)、jinfushanoside A(15)、cucurbitacine F(16)、16-O-acetyl-cucurbitacin F(17)。结论化合物1为新化合物,命名为彭县雪胆萜A;化合物2~9为首次从该属植物中分离得到,化合物1、5、10、14、16~17对He La细胞具有良好的抗肿瘤活性。Objective To investigate the chemical constituents of Hemsleya pengxianensis. Methods The compounds were separated and purified by silica gel column chromatography and semi-preparative high performance liquid chromatography(HPLC)methods, and their structures were identified by physicochemical property and NMR, IR, MS spectroscopic methods. The cytotoxic activity of the compounds against HeLa cells was determined by MTT assay. Results A total of 17 compounds were isolated and identified as hemsleyacin A(1), Jinfushanencin B(2), delavanoside D(3), scandenogenin D(4), jinfushanencin C(5), jinfushanoside D(6), hexanorcucurbitacin F(7), delavanoside A(8), cucurbitacin V(9), hemslepenside A(10), jinfushanencin F(11), scandenoside R3(12), scandenoside R2(13), hemslecin G(14), jinfushanoside A(15), cucurbitacine F(16), 16-O-acetyl-cucurbitacin F(17).Conclusion Compound 1 is a new type of cucurbitane triterpene, and compounds 2—9 are isolated from the Hemsleya genus for the first time.All of isolates were detected their inhibitory activities against HeLa cell lines by the MTT assay. The result showed that compounds 1, 5, 10, 14 and 16—17 exhibited inhibitory activity against the tested cell line.
