基于皮肤含水量、经皮失水量及血清VEGF含量探讨三黄消痤面膜对痤疮的治疗作用与机制

摘　　要：目的:探讨三黄消痤面膜对痤疮模型大鼠皮肤含水量、失水量及VEGF的干预作用以及可能的作用机制。方法:将48只SPF级大鼠随机分为空白组、模型组、三黄消痤面膜低剂量组(低剂量组)、三黄消痤面膜中剂量组(中剂量组)、三黄消痤面膜高剂量组(高剂量组)和维A酸乳膏组,每组8只。低、中、高剂量组给药剂量分别为0.05、0.1、0.2 g/(kg·d),维A酸乳膏组给药剂量为5 g/(kg·d)。除空白组外,其余各组大鼠涂抹100%油酸进行痤疮造模。造模3周结束后,各组大鼠(除空白组、模型组)分别给予相应剂量的药物治疗。于疗程结束后6、24、48 h和7、14 d分别测试大鼠皮肤含水量和经皮失水量。取鼠耳进行HE染色及形态学、病理学观察。末次治疗24 h后采用ELISA法检测大鼠血清VEGF含量变化。结果:治疗后,与维A酸乳膏组相比,中、高剂量组大鼠耳廓皮肤含水量、经皮失水量明显下降(P<0.05),血清中VEGF含量明显下降(P<0.05)。结论:三黄消痤面膜对痤疮有较好的治疗作用,其机制可能与促进痤疮大鼠皮肤屏障功能修复,调节大鼠血清中VEGF含量有关。Objective:To explore the effect of Sanhuang Xiaocuo Mask on water content,transdermal water loss and serum content of VEGF in acne model rats and to find out its possible mechanism in the treatment of acne.Methods:48 SPF rats were randomly divided into the blank group,the model group,the Sanhuang Xiaocuo Mask groups of low-dose[0.05 g/(kg·d)],medium-dose[0.1 g/(kg·d)]and high-dose[0.2 g/(kg·d)],as well as the Retinoic Acid Cream group[5 g/(kg·d)],with 8 rats in each group.The acne model was established by oleic acid applying to auricle duct opening.After 3 weeks of modeling,the rats in each group were given corresponding interventions.The skin water content and transdermal water loss were measured 6 h,24 h,48 h,7 d and 14 d after the treatment.The pathological changes were observed by HE staining method.The serum content of VEGF was measured by ELISA 24 h after the last treatment.Results:The water content was significantly increased and the amount of transdermal water loss was significantly decreased(P<0.05),and the serum content of VEGF was lowered in the Sanhuang Xiaocuo Mask groups of medium-dose and high-dose compared to those in the Retinoic Acid Cream group(P<0.05).Conclusion:Sanhuang Xiaocuo Mask has a good curative effect for acne,which mechanism may be related to promoting the repair of skin barrier and regulating the serum content of VEGF in rats with acne.
