南葶苈子水提液治疗肺源性心脏病合并心力衰竭模型大鼠的作用机制与研究

摘　　要：目的探讨南葶苈子水提液对肺源性心脏病合并心力衰竭大鼠的疗效及其作用机制。方法24只SD大鼠随机分为3组,即正常组(n=8)、模型组(n=8)和南葶苈子水提液组(n=8)。采用腹主动脉截面积缩窄术建立大鼠肺源性心脏病合并心力衰竭模型(正常组除外),灌胃给予药物处理。4周后用超声心动图检测心脏功能、肺质量指数以及尿量分析、肺组织苏木精-伊红(hematoxylin eosin,HE)染色、肺组织水通道蛋白1(aquaporins1,AQP1)以及心脏细胞凋亡和自噬状态分析。结果与模型组相比,南葶苈子水提液能明显改善肺源性心脏病合并心力衰竭大鼠减少左室后壁舒张末期厚度(LVPWd)(P<0.01),增加左室射血分数(LVEF)和左室短轴缩短率(FS)(P<0.01)。南葶苈子水提液可以显著增加大鼠5 h累积尿量(P<0.01),减少心力衰竭大鼠肺质量指数(P<0.01);肺脏HE染色显示南葶苈子水提液显著改善肺源性心脏病合并心力衰竭模型大鼠的肺水肿、肺部炎症细胞浸润等炎性状态;最后,免疫蛋白印迹提示南葶苈子水提液可以上调心力衰竭大鼠肺组织中的AQP1的表达(P<0.01),以及下调心脏凋亡相关蛋白caspase-3的表达(P<0.01),同时稳定心肌细胞自噬相关蛋白LC3B和p62的表达。结论南葶苈子水提液具有改善肺源性心脏病合并心力衰竭大鼠症状的功效,其发挥作用的途径可能是上调肺组织AQP1的表达,以及稳定心脏细胞凋亡和自噬状态,维持心肌细胞的功能。Objective To investigate the effect and mechanism of aqueous extract from Descurainiae Semen in rats with pulmonary heart disease combined with heart failure.Methods Total 24 SD rats were randomly divided into 3 groups:normal group(n=8),model group(n=8)and aqueous extract from Descurainiae Semen group(n=8).The model of pulmonary heart disease combined with heart failure(except the normal group)was established by abdominal aorta cross-sectional area constriction.Drugs were administered by gavage.Four weeks later,cardiac function was detected by echocardiography;lung mass index,urine volume,hematoxylin eosin(HE)staining in lung tissue,aquaporin 1(AQP1)in lung tissue,apoptosis and autophagy status of cardiac cells were analyzed.Results Compared with model group,aqueous extract from Descurainiae Semen significantly improved left ventricular posterior wall end-diastolic thickness(LVPWd)(P<0.01),increased left ventricular ejection fraction(LVEF)and shorted fraction shortening rate(FS)(P<0.01)in rats with pulmonary heart disease combined with heart failure.Aqueous extract from Descurainiae Semen significantly increased urine volume in 5 h(P<0.01),and reduced lung mass index(P<0.01)in rats with heart failure.HE staining showed that aqueous extract significantly improved pulmonary edema,lung inflammatory cell infiltration and other inflammatory states in rats with pulmonary heart disease combined with heart failure.Finally,Western blot indicated that aqueous extract from Descurainiae Semen up-regulated AQP1 expression(P<0.01)in lung tissues of rats with heart failure and down-regulated the cardiac apoptosis-related protein caspase-3 expression(P<0.01),stabilized the expression of autophagy related proteins LC3B and p62.Conclusion Aqueous extract from Descurainiae Semen has the effect of improving the symptoms of pulmonary heart disease combined with heart failure in rats by upregulating the expression of AQP1 in lung tissue,stabilizing the apoptosis and autophagy of heart cells,and maintaining the function of myocardial c
