摘  要:目的探讨七氟醚对脑缺血再灌注损伤(IRI)大鼠海马神经元三磷酸腺苷(ATP)酶活性的影响,并分析其机制。方法将30只雄性SD大鼠随机分为3组:正常组,模型组(建立局灶性脑IRI大鼠模型)和七氟醚组(脑IRI模型+七氟醚吸入治疗),每组各10只。电镜观察各组大鼠海马组织的超微结构,免疫荧光检测海马神经元含量,免疫组化检测海马环磷酸腺苷(cAMP)和蛋白激酶A(PKA)蛋白表达,试剂盒法检测海马ATP酶活性,荧光定量-聚合酶链锁反应(RT-PCR)和蛋白免疫印迹法(Western Blot)检测海马cAMP、PKA、环磷腺苷效应元件结合蛋白(CREB)和脑源性神经营养因子(BDNF)mRNA和蛋白表达。结果正常组海马神经元细胞膜结构完整,模型组海马神经元胞体萎缩,染色质浓缩并聚集在细胞核边缘,七氟醚组海马神经元形态明显改善。与对照组相比,模型组大鼠海马组织微管相关蛋白-2(MAP-2)阳性细胞占比显著降低(P<0.05),七氟醚组显著高于模型组(P<0.05)。与对照组相比,模型组大鼠海马组织cAMP和PKA阳性表达、Na^(+)-K^(+)-ATP酶和Ca^(2+)-ATP酶活性显著降低(P<0.05),七氟醚组显著高于模型组(P<0.05)。与对照组相比,模型组大鼠海马组织cAMP、PKA、CREB、BDNF mRNA和蛋白表达显著降低(P<0.05),七氟醚组显著高于模型组(P<0.05)。结论七氟醚可通过激活cAMP-PKA-CREB-BDNF信号通路来促进脑IRI大鼠海马神经元的ATP酶活性,进一步改善脑IRI。Objective To explore the effect of sevoflurane on the ATPase activity of hippocampal neurons in rats with cerebral ischemia reperfusion injury(IRI),and to analyze its mechanism.Methods Thirty male SD rats were ran domly divided into three groups:normal group,model group(focal cerebral IRI model)and sevoflurane group(cerebral IRI model+sevoflurane inhalation treatment),with 10 rats in each group.The ultrastructure of hippocampal tissue in each group was observed by electron microscopy,the content of hippocampal neurons was detected by immunofluorescence,the expression of cAMP and protein kinase A(PKA)in hippocampus were detected by immunohistochemistry,and the activity of hippocampal ATPase was detected by kit method.The mRNA and protein expressions of cAMP,PKA,cAMP-response element binding protein(CREB)and brain-derived neurotrophic factor(BDNF)in hippocampus were detected by quantitative real-time polymerase chain reaction(RT-PCR)and Western Blot.Results Hippocampal neurons in the normal group had intact cell membrane structure,while hippocampal neurons in the model group had atrophy,chromatin concentration and aggregation at the edge of nucleus,and hippocampal neurons in the sevoflurane group had significantly improved morphology.Compared with the control group,the proportion of MAP-2 positive cells in the model group was significantly decreased(P<0.05),and the sevoflurane group was significantly higher than the model group(P<0.05).Compared with the control group,the positive expressions of cAMP and PKA and the activities of Na^(+)-K^(+)-ATPase and Ca^(2+)-ATpase in the hippocampus of the model group were significantly decreased(P<0.05),and the sevoflurane group was significantly higher than the model group(P<0.05).Compared with the control group,the mRNA and protein expressions of cAMP,PKA,CREB and BDNF in the hippocampus of model group were significantly decreased(P<0.05),and those in the sevoflurane group were significantly higher than those in the model group(P<0.05).Conclusion Sevoflurane can promote the A
