摘  要:在不同环境下,细胞需要协调多种信号通路而有组织地适应不断变化的内外环境,以维持细胞稳态。鉴于疾病的复杂性,需要我们向整体研究的思路转变,以重要信号通路为立足点,深入了解疾病背景下信号通路在维系细胞稳态中的调控机制,为药物靶向治疗提供更多的理论基础。Phosphatidylinositiol 3-kinase(PI3 K)/protein kinase B(PKB/AKT)/mammalian target of rapamycin(mTOR) signaling pathway is a key signaling pathway that regulates cell growth,proliferation,differentiation,apoptosis and metabolism.It has been found that this signaling pathway is closely related to the occurrence and development of cancer,neurodegenerative diseases and many other diseases.Although targeting PI3 K/AKT/mTOR signaling pathway has been applied in anti-tumor therapy,increasing studies have found that the inhibitors of this signaling pathway up-regulate mitochondrial function through reprograming to reduce their anti-tumor effects.On the other hand,abnormal activation of PI3 K/AKT/mTOR signaling pathway during aging may lead to decreased mitochondrial homeostasis and induce cellular senescence.Therefore,the regulatory mechanism of mitochondrial homeostasis is an important factor to influence the effect of PI3 K/AKT/mTOR signaling pathway-targeted therapy on the diseases.In this article,we review the current knowledge on the roles of PI3 K/AKT/mTOR signaling pathway and their targets in some diseases,focusing on the interaction between PI3 K/AKT/mTOR signaling pathway and mitochondrial homeostasis.
