摘 要:通过筛选蛋鸡不同产蛋期卵巢组织差异表达基因(differentially expressed gene,DEGs)寻找影响蛋鸡繁殖性能的关键基因。试验提取5月龄(n=3)、6月龄(n=3)京红蛋鸡卵巢组织总RNA,构建测序文库,利用RNA-seq技术及生物信息学分析方法鉴定DEGs,并进行qRT-PCR验证和功能富集分析。结果表明,在不同月龄蛋鸡卵巢组织中共筛选鉴定到19586个蛋白质编码基因,其中63个存在差异表达,qRT-PCR验证了测序结果。对DEGs的功能富集显示其显著富集到273条GO条目及2条KEGG通路中,参与调控与蛋鸡繁殖性能相关的生物学过程,细胞组分,分子功能以及多种信号通路。研究结果表明,DEGs参与了能量代谢、物质合成、氧化应激等多个生物学途径。To identify the DEGs in different laying period and explore their relationship with production traits of laying hens,the total RNA were extracted from JingHong laying hens in different laying period(5 months age n=3 and 6 months age n=3).Then RNA-seq were carried out to identify DEGs.qRT-PCR was used to verify RNA-seq result and to predict the function of enrichment.A total of 19586 genes expressed in different ovarian tissues were identi fied,among them,63 genes were differentially expressed.qRT-PCR verified the sequencing results.Functional enrichment of differential expression of lncRNA showed significant enrichment in 273 GO entries and 2 KEGG pathways,involved in regulating biological processes,cell components,molecular function,and multiple signaling pathways.This study showed that DEGs in ovarian of laying hens in different laying period participates in different biological processes such as energy metabolism,material synthesis,and oxidative stress.
