解毒生肌膏对糖尿病溃疡大鼠创面局部IL-6、TNF-α表达的影响

摘　　要：目的:探究解毒生肌膏对2型糖尿病大鼠创面皮肤的组织形态学与白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)表达的影响。方法:采用链脲佐菌素(STZ)诱导SD雄性糖尿病大鼠,构建大鼠背部全层皮肤缺损模型并分为3组,解毒生肌膏组在皮肤创面造成后加中药解毒生肌膏干预,磺胺嘧啶银乳膏组加磺胺嘧啶银乳膏组干预,模型组不予处理,分别在第3天、第7天、第21天肉眼观察创面大体形态学,苏木精-伊红(HE)染色观察组织形态学,免疫组化检测炎症因子IL-6、TNF-α的阳性表达量。结果:与模型组比较,解毒生肌膏组大鼠创面局部经该药干预后创面愈合速度明显加快,愈合面瘢痕较少,炎症细胞数量减少,肉芽组织数量、成纤维细胞和胶原比例增多,IL-6和TNF-α含量均较低且峰值时间提前。结论:解毒生肌膏能够加快糖尿病皮肤创面愈合速度,提高愈合质量,其作用机理可能与降低炎症因子IL-6和TNF-α表达有关。Objective:To explore the effect of Jiedu Shengji Ointment on the histomorphology and expression of Interleukin-6(IL-6)and Tumor Necrosis Factor-α(TNF-α)in wounds of type 2 diabetic rats.Methods:Three groups of SD male diabetic rats induced by Streptozotocin(STZ)were used to construct full-thickness skin defect models respectively.The group of Jiedu Shengji Ointment was treated with Chinese medicine Jiedu Shengji Ointment after the skin wound was caused,the group of silver sulfadiazine cream was treated with the silver sulfadiazine cream,and the model group was not processed.On the 3rd,7th and 21st day of the experiment,gross morphology of the wound was observed with the naked eye,histomy was observed with Hematoxylin Yihong(HE)staining,and the positive expressions of inflammatory factors IL-6 and TNF-αwere detected by immunohistochemistry.Results:Compared with the model group,after the intervention of the drug,the wound healing speed of the rats in the Jiedu Shengji Ointment group was significantly accelerated with fewer scars on the healing surface,the number of inflammatory cells was reduced,the number of granulation tissue,fibroblasts and collagen were increased.Meanwhile,the content of IL-6 and TNF-αwas lower,and the peak time was earlier.Conclusion:Jiedu Shengji Ointment can speed up the healing of diabetic skin wounds and improve the quality of healing,which may be related to the reduction of the expression of inflammatory factors IL-6 and TNF-α.
