当归逐瘀胶囊对脑缺血再灌注不同时间点大鼠大脑皮层组织中p38、NF-κB65和STAT3蛋白的影响

摘　　要：目的探讨当归逐瘀胶囊对大鼠大脑中动脉闭塞/再灌注(MCAO/R)不同时间点皮层组织中p38蛋白(p38)、核转录因子-κB65(NF-κB65)和信号转导和转录激活因子3(STAT3)蛋白的影响。方法采用线栓法制备大鼠MCAO/R模型,缺血2 h待大鼠清醒后灌胃给药,假手术组与模型组灌胃给予蒸馏水10 mL/kg,当归逐瘀组灌胃给药2.00 g/kg,每天1次。再灌注24 h、48 h、72 h和7 d分别处死大鼠,取脑组织TTC染色法测梗死面积,干湿重量法测定脑含水量,HE染色观察组织病理学变化,Western Blot检测损伤侧脑组织p38、p-p38、NF-κB65、p-NF-κB65和STAT3蛋白表达。结果当归逐瘀能显著降低再灌注各时间点脑梗死面积(P<0.05);降低再灌注48 h、72 h、7 d脑组织含水量(P<0.05);有效改善大脑皮层病理组织损伤;再灌注24 h时,当归逐瘀胶囊能显著降低p-NF-κB65蛋白表达(P<0.05);再灌注48 h、72 h、7 d时可下调p-p38、p-NF-κB65和STAT3蛋白表达(P<0.05)。结论当归逐瘀胶囊有脑保护作用,其作用机制可能是通过调控p-p38、p-NF-κB65和STAT3蛋白的表达、抑制炎症反应而发挥脑保护作用。Objective To investigate the effect of Danggui Zhuyu Capsule on p38,NF-κB65 and STAT3 protein in cerebral cortex of rats with cerebral ischemia reperfusion at different time points.Methods MCAO/R model was established by thread suppository method.After ischemia for 2 hours,rats were given intragastric administration after waking up,the sham operation group and model group were given intragastric administration of distilled water 10 ml/kg,and the Danggui Zhuyu group were given intragastric administration of 2.00 g/kg,once a day.The rats were killed at 24 h,48 h,72 h and 7 d after reperfusion.The infarct size was measured by TTC staining,the cerebral edema was measured with dry-wet weight,the histopathological changes were observed by HE staining,and the expressions of p38,p-p38,NF-κB65,p-NF-κB65 and STAT3 protein in the injured brain were detected by Western blot.Results Compared with the model group,Danggui Zhuyu Capsule could significantly reduce the cerebral infarction area at each time point of cerebral ischemia-reperfusion(P<0.05);reduce cerebral edema content at 48 h,72 h and 7 d of reperfusion(P<0.05);effectively improve the pathological tissue injury of cerebral cortex;significantly reduce the protein expression of p-NF-κB65 at 24 h of reperfusion(P<0.05);and down regulate the protein expression of p-p38,p-NF-κB65 and STAT3 at 48 h,72 h and 7 d of reperfusion(P<0.05).Conclusion Danggui Zhuyu Capsule has the effect of anti-cerebral ischemia/reperfusion injury,the mechanisms may be related to reduce the expression of p-p38,p-NF-κB65 and STAT3,and inhibit inflammatory reaction.
