摘 要:目的研究番泻叶致幼龄大鼠腹泻模型量-时-效关系。方法将96只幼龄大鼠随机抽取24只作为空白组,余72只随机分为低、中、高浓度组,分别给予相应浓度番泻叶灌胃4周,建立大鼠腹泻模型。观察各组大鼠每周稀便率、腹泻指数、体重及电解质(Na+、K^(+)、CL^(-))变化。结果(1)腹泻指标:中、高浓度组每周稀便率、腹泻指数均显著高于低浓度组(P<0.01),中、高浓度第2、3、4周稀便率、腹泻指数高于第1周(P<0.05),高浓度组第4周稀便率显著高于第1周(P<0.01)。(2)体重:中、高浓度组每周体重增长较空白组、低浓度组缓慢(P<0.05),而高浓度组差异有显著意义(P<0.01)。(3)电解质:Na+:第2周中、高浓度组与空白组及高浓度组与低浓度组比较,下降有统计学意义(P<0.05);第3、4周中、高浓度组较空白组、低浓度组下降显著(P<0.01);中、高浓度组2、3、4周较第1周显著下降(P<0.01),而高浓度组第3、4周较第2周显著(P<0.01);K^(+):第2、3、4周中、高浓度组较空白组、低浓度组下降均有差异(P<0.05);中、高浓度组第2、3、4周较第1周下降均有差异(P<0.05),而高浓度组下降显著(P<0.01);CL^(-):第2周中、高浓度组较空白组、低浓度组下降有统计学意义(P<0.05),而第3、4周下降显著(P<0.01)。中、高浓度组第2周较第1周下降有统计学意义(P<0.05),而第3、4周下降显著(P<0.01);(4)安全性:高浓度组较中浓度组死亡率高,差异有统计学意义(P<0.05)。(5)结肠病理:中、高浓度组第3、4周较第1、2周相对较重,但差异无统计学意义(P>0.05)。结论中浓度番泻叶致幼龄大鼠腹泻2周时模型效果理想,安全性好。Objective To study the dose-time-effect relationship of diarrhea model in young rats induced by Folium sennae.Methods 24 rats were randomly selected from 96 young rats as the blank group,and 72 rats were randomly divided into the low,medium and high concentration groups.The rats were given Folium sennae with different concentrations by gavage for 4 weeks,to establish the rat diarrhea model.The changes of loose stools,diarrhea index,weight and electrolyteswere observed in each group.Results(1)Diarrhea:The weekly rate of loose stools and diarrhea index of the medium and high concentration groups were significantly higher than the low concentration groups(P<0.01).At week 2,3 and 4,the rate of loose stools and diarrhea index of the medium and high concentration groups were higher than week 1(P<0.05).At week 4,the rate of loose stools of the high concentration groups were significantly higher than week1(P<0.01).(2)Weight:The weekly weight of the medium and high concentration groups were slower than the blank groups and low concentration groups(P<0.05),the weight of the high concentration groups were significantly slower(P<0.01).(3)Electrolyte:Na;:At week 2,the levels of the medium and high concentration groups were lower than the blank groups,while those of the high concentration groups were lower than the low concentration groups(P<0.05),at week 3 and 4,the levels of the medium and high concentration groups were significantly lower than the blank and low concentration groups(P<0.01),the levels of the middle and high concentration groups were significantly lower at week 2,3 and 4 than at week 1(P<0.01),the levels of the high concentration groups were significantly lower at week 3 and 4 than at week 2(P<0.01).K^(+):At week 2,3,and 4,the levels of the medium and high concentration groups were lower than the blank and low concentration groups(P<0.05),at week 2,3,and 4,the levels of the medium and high concentration groups were lower with week 1(P<0.05),the levels of high concentration groups dropped significantly(P<0.01)
