摘 要:目的观察寒凝血瘀型EMs大鼠模型腹腔液中AQP1、AQP2、AQP5的mRNA表达,并观察少腹逐瘀汤对其表达水平的影响。方法选用195只雌性未交配Wistar大鼠,随机分为空白组、模型组、少腹逐瘀汤低剂量组、少腹逐瘀汤中剂量组、少腹逐瘀汤高剂量组、阳性对照组(达那唑)6组,除空白组外,其余各组用冰水浸泡及自体移植结合法建立寒凝血瘀型EMs大鼠模型,采用RT-PCR方法测定各组大鼠腹腔液内AQP1、AQP2、AQP5的mRNA表达水平。结果与空白对照组比较,模型组大鼠腹腔液AQP1、AQP5的mRNA含量均升高,差异显著(P<0.01),AQP2的mRNA含量下降,差异具有统计学意义(P<0.05)。经少腹逐瘀汤干预模型大鼠后,与模型组对比,少腹逐瘀汤各剂量组AQP1、AQP5的mRNA含量均明显降低,差异具有统计学意义(P<0.05),AQP2的mRNA含量明显升高,差异显著(P<0.01)。结论EMs存在腹腔液内水通道蛋白的表达的异常,少腹逐瘀汤可以调节EMs腹腔液当中的水通道蛋白的mRNA表达。Objective To observe the mRNA expressions of AQP1,AQP2 and AQP5 in the abdominal fluid of EMs rats with cold coagulation and blood stasis,and to observe the effect of Shaofu Zhuyu Decoction on its expression level.Methods Choose 195 unmated female Wistar rats were randomly divided into blank group,model group,low dose group,the lower abdomen by stasis soup in the lower abdomen by stasis soup dose group,high dose group,the lower abdomen by stasis soup(danazol)6 group,positive control group except the blank group,the rest of the group and in combination with autologous transplantation in an ice water EMs haemorrheological nature blood stasis syndrome rats model,using rt-pcr method to determine each rat peritoneal fluid inside AQP1,AQP2 and AQP5 mRNA expression level.Results Compared with the blank control group,the mRNA levels of AQP1 and AQP5 in the abdominal fluid of the model group were increased,with significant differences(P<0.01),while the mRNA levels of AQP2 were decreased,with statistically significant differences(P<0.05).After the intervention of Shaofu Zhuyu Decoction,the mRNA contents of AQP1 and AQP5 in each dose group were significantly reduced compared with the model group(P<0.05),and the mRNA contents of AQP2 were significantly increased(P<0.01).Conclusion EMs has abnormal expression of aquaporin in peritoneal fluid,and Shaofu Zhuyu Decoction can regulate mRNA expression of AQUaporin in EMs peritoneal fluid.
