摘 要:目的探讨miR-125b-5p对AngⅡ诱导的的心肌成纤维细胞(CFs)转分化的影响及抗纤益心方抑制心肌纤维化的作用机制。方法 CFs取于出生48 h内乳鼠,体外培养并分为正常组,模型组,抗纤益心方组。利用腺病毒转染miR-125b-5p, q-PCR检测miR-125b-5p以及-SMA mRNA表达水平;Western blot检测Col-1、-SMA蛋白表达水平。结果与正常组比较,模型组miR-125b-5p以及-SMA mRNA表达水平明显增加(P<0.05)。与模型组比较,抗纤益心方组miR-125b-5p和-SMA mRNA表达水平均下降(P<0.05),Col-1、-SMA蛋白表达水平降低;转染组-SMA表达降低(P<0.05)。结论抗纤益心方可能通过下调miR-125b-5p表达抑制扩张型心肌病心肌纤维化。Objective To explore the effect of miR-125 b-5 p on the transdifferentiation of cardiac fibroblasts(CFs)induced by AngⅡand the mechanism of Kangxian Yixin Decoction in inhibiting cardiac fibrosis.Methods CFs were taken from suckling mice within 48 hours of birth,cultured in vitro and divided into normal group,model group,and Kangxian Yixin Decoction group.Transfect miR-125 b-5 p with adenovirus.q-PCR was used to detect the expression levels of miR-125 b-5 p andα-SMA mRNA;Western blot was used to detect the expression levels of Col-1 andα-SMA proteins.Results Compared with the normal group,the expression levels of miR-125 b-5 p andα-SMA mRNA in the model group increased significantly(P<0.05).Compared with the model group,the expression levels of miR-125 b-5 p and-SMA mRNA in the Kangxian Yixin Decoction group decreased(P<0.05),and the expression levels of Col-1 andα-SMA proteins decreased;the expression ofα-SMA decreased in the transfection group(P<0.05).Conclusion Kangxian Yixin Decoction may inhibit dilated cardiomyopathy myocardial fibrosis by down-regulating the expression of miR-125 b-5 p.
