摘 要:目的:研究由金银花、黄连、甘草等组成的复方中草药制剂对青脚麻鸡胸腺显微结构、免疫功能以及凋亡相关基因Caspase-3 mRNA和蛋白表达的影响。方法:选用125羽健康28 d未接种法氏囊疫苗的青脚麻鸡,按照体重相近原则随机分对照组、攻毒组和试验Ⅰ~Ⅲ组,每组5个重复(每个重复5羽),对照组和攻毒组饲喂基础饲粮,试验Ⅰ~Ⅲ组分别添加0.5%、1.0%和1.5%的复方中草药制剂。攻毒组和试验组在30 d通过点眼人工感染IBDV,试验期为7 d。结果:与对照组相比,IBDV攻毒第3天,青脚麻鸡胸腺指数、Caspase-3 mRNA和蛋白相对表达量以及胸腺单位面积Caspase-3阳性细胞数量均显著升高(P<0.05),而胸腺皮质厚度和细胞因子(IL-2、INF-γ)水平均显著降低(P<0.05);相比攻毒组,试验Ⅱ组青脚麻鸡胸腺指数、Caspase-3 mRNA和蛋白相对表达量均显著降低(P<0.05),而皮质厚度和IL-2含量均显著升高(P<0.05)。IBDV攻毒第7天,与对照组相比,青脚麻鸡胸腺指数、Caspase-3 mRNA表达量和胸腺单位面积Caspase-3阳性细胞数显著升高(P<0.05),而胸腺皮质厚度和细胞因子(IL-2、INF-γ)水平均显著降低(P<0.05);相比攻毒组,试验Ⅱ组青脚麻鸡胸腺指数、Caspase-3 mRNA相对表达量和胸腺单位面积Caspase-3阳性细胞数均显著降低(P<0.05)。显微观测发现,IBDV攻毒第3天和第7天,与对照组相比,试验Ⅱ组青脚麻鸡胸腺显微结构无明显变化,皮质和髓质结构清晰,细胞排列紧密。结论:饲粮添加1.0%复方中草药制剂对IBDV感染青脚麻鸡胸腺显微结构有明显改善作用,并能提高胸腺免疫,抑制胸腺淋巴细胞凋亡,减轻IBDV对青脚麻鸡胸腺的损伤。Objective: To explore the effects of compound Chinese herbal medicine(such as honeysuckle, coptis, licorice, etc.) on microstructure, immune function of Partridge Shank chicken thymus and the expression of apoptosis-related gene Caspase-3 mRNA and protein. Methods: According to the principle of similar body weight, 125 healthy 28-day-old Partridge Shank chicken that were not vaccinated with IBD vaccine were randomly divided into control group, the challenge group and experiment group Ⅰ-Ⅲ, with 5 replicates in each group(5 per repeat). The control group and the challenge were fed on basal diets, and the experiment groups Ⅰ-Ⅲ were fed on 0.5%, 1.0% and 1.5% compound Chinese herbal medicine respectively. The challenge group and the experiment group were infected with IBDV through eye drops at the age of 30-day-old, and the experiment period was 7 d. Results: Compared with the control group after 3 days of IBDV challenge, the thymus index, relative expression of Caspase-3 mRNA and protein, and the number of Caspase-3 positive cells of Partridge Shank chicken of challenge group were significantly increased(P<0.05), but the thickness of thymus cortex, cytokine(IL-2, INF-γ) were significantly reduced(P<0.05). Compared with the challenge group, the thymus index and the relative expression of Caspase-3 mRNA and protein in experiment group II were significantly reduced(P<0.05), but the cortical thickness and IL-2 content were significantly increased(P<0.05). Compared with the control group after 7 days of challenge, the thymus index, Caspase-3 mRNA and Caspase-3 positive cells of thymus in the challenge group were significantly increased(P<0.05), but the cortical thickness and cytokine(IL-2, INF-γ) content were significantly reduced(P<0.05).
