摘 要:目的探讨去甲肾上腺素(NE)对急性心肌梗死(AMI)后小鼠左心室重构及心功能的影响。方法用结扎前降支结扎构建小鼠AMI模型,并随机均分为模型组及实验组,每组10只;另取10只小鼠不结扎前降支动脉作为假手术组。模型组和假手术组均给予腹腔注射等体积0.9%NaCl;实验组给予腹腔注射2 mg·kg^(-1) NE。3组小鼠每天给药1次,连续给药7 d。用超声心动图评估左心室重构和心功能,用酶联免疫吸附实验检测小鼠血清白细胞介素-6(IL-6)及肿瘤坏死因子-α(TNF-α)浓度。结果实验组、模型组和假手术组的左心室短轴缩短率分别为(35.36±6.26)%,(40.25±5.81)%和(48.14±7.43)%,左心室舒张末期内径分别为(4.57±1.18),(4.26±0.86)和(3.97±0.65)mm,左心室收缩末期内径分别为(2.93±0.70),(2.57±0.64)和(2.01±0.53)mm, IL-6分别为(345.86±114.69),(279.19±76.48)和(61.14±12.43)pg·mL^(-1),TNF-α分别为(83.65±18.87),(61.14±13.43)和(39.57±6.28)pg·mL^(-1),心脏指数分别为(6.31±1.42),(5.08±0.96)和(4.87±0.73)g·kg^(-1),实验组的上述指标与模型组和假手术组相比,差异均有统计学意义(均P<0.05)。结论 NE是诱导AMI后小鼠左心室重构的因素,可能与其加剧心肌炎症反应有关。Objective To investigate the effect of norepinephrine(NE)on the left ventricular remodeling and cardiac function in mice after acute myocardial infarction(AMI).Methods The mouse AMI model was constructed with pre-ligation descending ligation,and was randomly divided into model and experimental groups with 10 per group.Another10 mice without ligation of anterior descending artery were selected as the sham-operation group.The model and sham-operation groups received0.9%NaCl.The experimental group was given intraperitoneal injection of2 mg·kg^(-1) NE.Three groups were administered once a day for 7 days.Left ventricular remodeling and cardiac function were evaluated by echocardiography,and mouse serum interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)concentrations were detected by enzyme-linked immunosorbent assay.Results The short-axis shortening rates of the left ventricle in experimental,model and sham-op eration groups were(35.36±6.26)%,(40.25±5.81)%and(48.14±7.43)%,the end-diastolic diameters of left ventricle were(4.57±1.18),(4.26±0.86)and(3.97±0.65)mm,the left ventricular end-systolic diameters were(2.93±0.70),(2.57±0.64)and(2.01±0.53)mm,IL-6 were(345.86±114.69),(279.19±76.48)and(61.14±12.43)pg·mL-1,TNF-αwere(83.65±18.87),(61.14±13.43)and(39.57±6.28)pg·mL-1,heart indexes were(6.31±1.42),(5.08±0.96)and(4.87±0.73)g·kg^(-1).The above indexes The above indexes in the experimental group were statistically significant compared with the model group and the sham operation group(all P<0.05).Conclusion NE is a factor inducing left ventricular remodeling in mice after AMI,which may be associated with its aggravation of myocardial inflammation.
