摘 要:目的研究蒙药平喘灵胶囊对哮喘小鼠的作用机制。方法用鸡蛋清白蛋白建立哮喘小鼠模型。按照体重将雌性BALB/c小鼠随机分为6组:正常组、模型组、阳性对照组和高、中、低3个剂量实验组,每组10只。从造模持续第21天开始,高、中、低3个剂量实验组给予平喘灵1.32,0.66,0.33 g·kg^(-1),阳性对照组给予地塞米松片4.5 mg·kg^(-1)灌胃,正常组灌胃蒸馏水,连续给药8周。用细胞计数法计数支气管肺泡灌洗液中白细胞数,用蛋白质印迹法和荧光定量-PCR法检测肺组织中转录因子T-bet蛋白(灰度值)和基因(2^(-ΔΔCT)值)的表达水平。结果正常组、模型组、阳性对照组和高、中、低3个剂量实验组的支气管肺泡灌洗液中白细胞数量分别为13.90±1.10,116.70±8.20,57.27±2.71,78.36±1.54,87.91±1.61和95.15±2.11;转录因子T-bet蛋白的相对表达量分别为2.04±0.06,0.85±0.09,1.89±0.11,1.39±0.60,1.16±0.74和1.08±0.60;转录因子T-bet基因结果的趋势与蛋白一致。上述指标:模型组与正常组比较,差异均有统计学意义(均P<0.05);3个剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论蒙药平喘灵胶囊能防治小鼠哮喘,可能是通过转录因子T-bet/转录因子GATA-3通路发挥作用。Objective To investigate the effect and mechanism of Mongolian Medicine Pingchuanling Capsule(MMPC)on asthmatic mice.Methods The mice were randomly divided into six groups according to weight:Normal group,model group,positive control group and high dose,medium dose and low dose experimental groups.The ovalbumin(OVA)was used to establish the asthma mice model.From the 21 st day of modeling,the mice in experiment group were given MMPC 1.32,0.66,0.33 g·kg^(-1) at high,medium and low doses respectively;positive control group were given dexamethasone tablet 4.5 mg·kg^(-1) by intragastric administration.Saline was used to replace OVA in normal group,and distilled water by intragastric administration.Continuous administration of 8 weeks.The number of white blood cells in bronchoalveolar lavage fluid was counted by cell counting.The protein expression(gray value)of transcription factors T-box expressed in T cell(T-bet)in the lung tissue were evaluated by Western blot.The gene expression(2-^(ΔΔCT)value)of transcription factor T-bet mRNA in lung tissues was detected by fluorescence quantitative-PCR.Results The number of white blood cells in bronchoalveolar lavage fluid in normal group,model group,positive control group and high,medium and low doses experimantal groups were 13.90±1.10,116.70±8.20,57.27±2.71,78.36±1.54,87.91±1.61,95.15±2.11,respectively;the protein expression of transcription factors T-bet in the six groups were2.04±0.06,0.85±0.09,1.89±0.11,1.39±0.60,1.16±0.74,1.08±0.60,respectively.The trend of gene Results is consistent with that of protein.Comparison between model group and normal group,the difference of the factors were significant(all P<0.05);comparison between three doses experimental groups and model group,the difference of the factors were significant(all P<0.05).Conclusion MMPC can alleviate asthma in mice.It works may through transcription factors T-bet/GATA-3 pathway.
