摘 要:为探明鸭坦布苏病毒病重组亚单位疫苗的安全性、有效性和保护性,试验将550只7日龄樱桃谷雏鸭随机分成11组,50只/组,1~10组分别免疫不同剂量重组亚单位疫苗(3个批次,每个批次设低、中、高剂量组)、商品活疫苗,11组为非免疫对照组。采用ELISA方法检测鸭血清IgG抗体水平,用ELISA测定试剂盒测定鸭血清中白细胞介素4(IL-4)和γ干扰素(IFN-γ)含量,用MTT法测定T淋巴细胞增殖水平,并进行中和抗体试验、攻毒保护试验、安全性试验测定中和抗体效价,评价疫苗安全性和保护率。结果表明:3个批次重组亚单位疫苗均能刺激机体产生高水平抗体,且中剂量组的效果显著高于低剂量组(P<0.05),与高剂量组及商品活疫苗组差异不显著(P>0.05)。高剂量组免疫鸭体内均有IL-4和IFN-γ分泌,且含量与商品活疫苗组差异不显著(P>0.05),疫苗能刺激雏鸭T淋巴细胞增殖,中、高剂量组产生的中和抗体效价与商品活疫苗组差异不显著(P>0.05),非免疫对照组鸭均未产生IgG抗体与中和抗体,低剂量组的疫苗保护率为70%或80%,中、高剂量组与商品活疫苗组的疫苗保护率均为100%,非免疫对照组鸭均表现为典型的坦布苏病毒感染症状,其中4只死亡,免疫鸭食欲、精神状态均正常,注射部位未见明显病变,体重增长情况与非免疫对照组鸭一致。说明该鸭坦布苏病毒病重组亚单位疫苗具有良好的免疫效力、保护性及安全性。In order to explore the safety, efficacy and protection of a subunit vaccine of duck Tembusu virus disease, in the experiment, 550 7-day-old Cherry-Valley ducklings were randomly divided into 11 groups with 50 per group, and 1-10 groups were immunized with different doses of recombinant subunit vaccine immunization group(three batches with low, medium and high doses per batch),commercial live vaccine, and a non-immunization control group(11 group). ELISA method was used to detect the level of IgG antibody in duck serum, and the contents of IL-4 and IFN-γ in duck serum was determined by ELISA assay kit. MTT method was used to determine the proliferation level of lymphocytes, and neutralizing antibody test, challenge protection test, and safety test were conducted to determine the neutralizing antibody titer and to evaluate the vaccine protection rate. The results showed that three batches of recombinant subunit vaccines could all stimulate the body to produce high levels of antibodies, and the effect of the medium-dose group was significantly higher than that of the low-dose group(P<0.05),which was not significantly different from the high-dose group and the commercial live vaccine group(P>0.05). The immunized ducks both secreted IL-4 and IFN-γ,and the content was not significantly different from the commercial live vaccine group(P>0.05). The vaccine could stimulate T lymphocyte proliferation of ducklings. There was no significant difference in the level of neutralizing antibody produced by medium dose group and high dose group compared with commercial live vaccine group(P>0.05). None of the non-immunized control ducks produced IgG antibody and neutralizing antibody. The protection rate of low dose group was 70% or 80%,and that of medium dose group, high dose group and commercial live vaccine group was 100%. All the ducks in the non-immunization control group showed typical symptoms of Tembusu virus infection, and four of them died.
