摘  要:目的研究牡蛎肽(Oyster peptide,OP)对CCl4诱导肝纤维化大鼠核因子-κB(NF-κB)、诱导型一氧化氮合成酶(iNOS)的影响及相关机制。方法50只SD大鼠随机分组:正常组、模型组、OP低剂量组、OP中剂量组、OP高剂量组。采用免疫组化、RT-PCR和Western blot方法分别检测肝组织Ⅰ、Ⅲ型胶原、NF-κB、iNOS的表达。吖啶橙/溴化乙锭双重染色分析肝星状细胞(HSC)凋亡。结果(1)免疫组化结果显示:与模型组比较,正常组、OP干预组的Ⅰ、Ⅲ型胶原蛋白均减少,差异有统计学意义(P<0.05);OP低剂量组、OP中剂量组与OP高剂量组之间Ⅰ、Ⅲ型胶原表达两两比较差异有统计学意义(P<0.05);(2)模型组与OP低剂量组HSC凋亡率比较差异无统计学意义(P>0.05);经中高剂量OP干预后HSC凋亡率与其他各组比较明显升高,差异有统计学意义(P<0.05);(3)RT-PCR和Western blot结果显示:正常组仅少量NF-κB、iNOS表达,模型组NF-κB、iNOS表达明显增多,与正常组比较差异有统计学意义(P<0.05)。OP干预组NF-κB、iNOS表达较模型组明显减少,且随OP剂量增加NF-κB、iNOS表达逐渐减少,各干预组间比较差异均有统计学意义(P<0.05)。结论OP可以抑制大鼠肝纤维化,呈剂量依赖性,抗肝纤维化机制可能与抑制NF-κB/iNOS途径有关。Objective To study the expression and related mechanisms of oyster peptide(OP)on nuclear factor-κB and inducible nitric oxide synthase in rats with CCl4-induced liver fibrosis.Methods 50 SD rats were randomly divided into normal group,model group,OP low-dose group,OP medium-dose group,OP highdose group.Immunohistochemistry,RT-PCR and Western blot methods were used to detect the expression of typeⅠandⅢcollagen,nuclear factor-κB(NF-κB)and inducible nitric oxide synthase(iNOS)in liver tissue.Acridine orange/ethidium bromide double staining was used to analyze hepatic stellate cell(HSC)apoptosis.Results(1)The results of immunohistochemistry showed that compared with model group,typeⅠandⅢcollagen were reduced in OP intervention group and normal group,and the differences were statistically significant(P<0.05).There were differences in the expression of typeⅠandⅢcollagen among OP low-dose group,OP middle-dose group and OP high-dose group,and the pairwise comparisons were statistically significant(P<0.05).(2)There was no significant difference in the HSC apoptosis rate between model group and OP low-dose group(P>0.05).The HSC apoptosis rate of OP medium-dose group and OP high-dose group was significantly higher than that of the other groups,and the differences were statistically significant(P<0.05).(3)RT-PCR and Western blot results showed that only a small amout of NF-κB,iNOS were expressed in normal group,and the expression of those were significantly increased in model group,the difference was statistically significant(P<0.05).The expression of NF-κB and iNOS in OP intervention group were significantly lower than those in model group,and the expression of NF-κB and iNOS decreased gradually with the increase of OP dose.The difference among the intervention groups was statistically significant(P<0.05).Conclusion OP can inhibit liver fibrosis in rats in a dose-dependent manner.The mechanism of anti-liver fibrosis may be related to the inhibition of NF-κB/iNOS pathway.
