摘 要:目的:探讨广枣总黄酮(total flavnoids of fructus choerospondiatis,TFFC)对三氧化二砷(Arsenic trioxide,ATO)诱导大鼠心肌损伤的保护作用。方法:采用灌胃5mg·kg^(-1) ATO溶液建立动物损伤模型。实验设为空白组、模型组(ATO 5mg·kg^(-1))、TFFC低剂量组(TFFC 15mg·kg^(-1)+ATO 5mg·kg^(-1)),TFFC中剂量组(TFFC 30mg·kg^(-1)+ATO 5mg·kg^(-1))、TFFC高剂量组(60 mg·kg^(-1)+ATO 5mg·kg^(-1)),空白组大鼠灌胃给予相同体积的药物溶媒0.5%羧甲基纤维素钠(CMC-Na),连续灌胃2周。用药期间监测大鼠体重,2周后用10%水合氯醛将大鼠麻醉,用生物机能实验系统测定大鼠心电图变化;腹主动脉取血,测定大血清中磷酸肌酸激酶(CK)、天冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)的活力;处死大鼠取心脏称重,计算心体比;检测心肌组织中过氧化氢酶(CAT)、超氧化物歧化酶(SOD)的谷胱甘肽过氧化物酶(GSH-Px)的活力。结果:模型组大鼠体重及心体比显著降低(P<0.05),血清中CK、AST、LDH的活力显著升高(P<0.05),心肌组织中CAT、SOD、GSH-Px的活力显著下降(P<0.05);与模型组比较,TFFC各剂量组大鼠体重及心体比显著升高(P<0.05),血清中CK、AST、LDH的活力显著降低(P<0.05),心肌组织中CAT、SOD、GSH-Px的活力显著升高(P<0.05)。结论:TFFC对ATO诱导大鼠心肌损伤具有显著的保护作用。Objective:To explore the protective effect of total flavnoids of fructus choerospondiatis(TFFC)on arsenic trioxide(ATO)-induced myocardial injury in rats.Methods:The animal injury model was established by intragastric administration of 5 mg·kg^(-1) ATO solution.The experiment was set as blank group,model group(ATO 5mg·kg^(-1)),TFFC low-dosegroup(TFFC 15mg·kg^(-1)+ATO 5mg·kg^(-1)),TFFC middle-dose group(TFFC 30mg·kg^(-1)+ATO 5mg·kg^(-1)),TFFC high-dose group(60mg·kg^(-1)+ATO 5mg·kg^(-1)),rats in the blank group were given the same volume of drug vehicle 0.5%sodium carboxymethyl cellulose(CMC-Na),continuous gavage for 2 weeks.The body weight of the rats was monitored during the medication.After 2 weeks,the rats were anesthetized with 10%chloral hydrate,and the changes in the electrocardiogram of the rats were measured with the biological function experiment system;Blood was taken from the abdominal aorta to determine the serum phosphocreatine kinase(CK),the activity of aspartate aminotransferase(AST)and lactate dehydrogenase(LDH);The rats were sacrificed and the hearts were weighed,and the heart-body ratio was calculated;and the detection of catalase(CAT)and superoxide dismutation in myocardial tissue enzyme(SOD)activity of glutathione peroxidase(GSH-Px).Results:The body weight and heart-body ratio of the model group were significantly reduced(P<0.05),the activities of CK,AST,and LDH in the serum were significantly increased(P<0.05),and the activities of CAT,SOD,and GSH-Px in the myocardial tissue were significant decreased(P<0.05);Compared with the model group,the body weight and heart-body ratio of rats in each dose of TFFC were increased significantly(P<0.05),the activities of CK,AST,and LDH in the serum were significantly decreased(P<0.05).The vitality of CAT,SOD and GSH-Px in the tissues were significantly increased(P<0.05).Conclusion:TFFC has a significant protective effect on ATO-induced myocardial injury in rats.
