摘  要:目的观察氟西汀对单眼形觉剥夺性弱视大鼠视觉功能及视皮质突触可塑性的影响,探讨其可能机制。方法SD大鼠45只,随机选取10只为正常组,不作处理,余35只行左侧眼睑缝合术制备单眼形觉剥夺性弱视大鼠模型。将造模成功30只大鼠随机分为模型组、氟西汀组、氟西汀+舒林酸组各10只。氟西汀组腹腔注射氟西汀生理盐水溶液10 mg/kg,氟西汀+舒林酸组腹腔注射氟西汀生理盐水溶液10 mg/kg+舒林酸生理盐水溶液5 mg/kg,模型组、正常组腹腔注射等量生理盐水,均1次/d,持续2周。给药2周后测定4组大鼠剥夺眼闪光视觉诱发电位,记录P波潜伏期及振幅。4组大鼠处死后取剥夺眼侧视皮质,测定视皮质突触数量、突触间隙宽度、突触后致密带厚度;行HE染色观察视皮质组织形态;采用Western blot法检测视皮质糖原合成酶激酶-3β(glycogen synthase kinase-3β,GSK-3β)、p-β-catenin、β-catenin蛋白相对表达量,计算p-β-catenin/β-catenin。结果模型组剥夺眼P波潜伏期[(113.35±12.64)ms]长于正常组[(77.62±11.57)ms](P<0.05),振幅[(5.66±1.47)μV]低于正常组[(21.53±3.61)μV](P<0.05),视皮质突触数量[(39.80±5.07)个/200μm^(2)]少于正常组[(61.60±8.53)个/200μm^(2)](P<0.05),突触间隙宽度[(26.37±3.18)nm]和突触后致密带厚度[(77.01±7.76)nm]均大于正常组[(15.56±2.37)、(49.93±5.14)nm](P<0.05);氟西汀组剥夺眼P波潜伏期[(87.81±14.53)ms]短于模型组(P<0.05),振幅[(14.89±2.73)μV]高于模型组(P<0.05),视皮质突触数量[(53.20±6.14)个/200μm^(2)]多于模型组(P<0.05),突触间隙宽度[(18.34±2.88)nm]和突触后致密带厚度[(59.08±6.03)nm]均小于模型组(P<0.05);氟西汀+舒林酸组剥夺眼P波潜伏期[(104.23±13.61)ms]长于氟西汀组(P<0.05),振幅[(10.17±1.95)μV]低于氟西汀组(P<0.05),视皮质突触数量[(45.80±5.89)个/200μm^(2)]少于氟西汀组(P<0.05),突触间隙宽度[(21.19±3.07)nm]和突触后致密带厚度[(65.42±7.15)Objective To observe the influence of fluoxetine on visual function and synaptic plasticity of optic cortex neurons in monocular form-deprived amblyopic rats,and to explore its potential mechanisms.Methods In 45 SD rats,10 were randomly selected as normal group without treatment,and the remaining 35 underwent left eyelid suture to establish monocular form-deprived amblyopic rats models.Thirty rats were successfully modeled,and were randomly divided into model group,fluoxetine group,fluoxetine+sulindac group,with 10 rats in each group.After successfully modeling,fluoxetine group was intraperitoneally injected with fluoxetine 10 mg/kg,fluoxetine+sulindac group was intraperitoneally injected with fluoxetine 10 mg/kg and sulindac 5 mg/kg,normal group and model group were intraperitoneally injected with the same amount of normal saline,once a day totally for two weeks.After two weeks of administration,the flash visual evoked potential of the affected eye in each group was measured,and the P wave latency and amplitude were recorded.After the rats of each group were sacrificed,the brain tissue of the visual cortex of the affected eye was taken,and the number of synapses of the visual cortex neurons,the width of the synaptic gap,and the thickness of the post-synaptic dense zone were measured.The morphology of visual cortex was observed by HE staining.The relative expressions of glycogen synthase kinase-3β(GSK-3β),β-catenin,and p-β-catenin protein in visual cortex were detected by Western blot.Results The P wave latency was longer in model group[(113.35±12.64)ms]than that in normal group[(77.62±11.57)ms](P<0.05),the amplitude was lower in model group[(5.66±1.47)μV]than that in normal group[(21.53±3.61)μV](P<0.05),the number of synapses was less in model group[(39.80±5.07)/200μm^(2)]than that in normal group[(61.60±8.53)/200μm^(2)](P<0.05),and the width of the synaptic gap and post-synaptic dense zone thickness were greater in model group[(26.37±3.18),(77.01±7.76)nm]than those in normal group[(15.56±2.37),
