摘  要:为了探究海南黑山羊生长分化因子9(GDF9)和骨形态发生蛋白15(BMP15)基因多态性与初胎产羔数间的关系,采用PCR扩增海南黑山羊GDF9和BMP15基因序列,通过基因测序检测2个基因中SNP位点的多态性分布情况,并与初胎产羔数进行关联分析。结果:在海南黑山羊GDF9基因中共发现3处碱基突变,分别位于GDF9基因的第1外显子、第2外显子和3′端,在BMP15基因中共发现2处碱基突变,分别位于BMP15基因的5′端和第2外显子;在GDF9基因+183处的A/C突变位点、+2 082处的A/C突变位点、+2 541处的C/T突变位点上,海南黑山羊的优势基因型分别是CC、AA和CT,在BMP15基因-519处的A/G突变位点、+6 124处的C/G突变上,海南黑山羊的优势基因型分别是AG和CC;GDF9基因+2 541处的C/T突变与初胎产羔数呈显著相关,TT基因型个体的产羔数显著高于CC基因型个体(P<0.05)。本试验为通过基因标记辅助选择提高海南黑山羊的产羔数奠定基础。This study was to explore the effects of the polymorphisms of growth differentiation factor 9(GDF9) and bone morphogenetic protein 15(BMP15) on the litter size of the first parity of Hainan black goats. GDF9 and BMP15 genes were amplified by PCR. The polymorphisms of the two genes were identified by sequencing. Then, correlation between the polymorphisms of GDF9 and BMP15 genes with the litter size of the first parity of the tested goats was analyzed by the SAS statistic software. The results showed that three mutations were detected in the GDF9 genomic fragment, one in the first exon, one in the second exon and one in the 3′ regulatory region. A total of 2 mutations were detected in the BMP15 genomic fragment, one in the 5′ regulatory region and one in the second exon. At the A+183 C locus, A+2 082 C locus, C+2 541 T locus of the GDF9 gene, the CC, AA and CT genotypes were predominant in Hainan black goats, respectively. At the A-519 G locus and C+6 124 G locus of the BMP15 gene, the AG and CC genotypes were predominant in the black goats, respectively. Association analyses revealed that the C+2 541 T locus of the GDF9 gene was significantly associated with the litter size of the first parity of the goats. The litter size of the goats with the TT genotype was significantly higher than that of the goats with the CC genotype(P<0.05). These results provide scientific bases for improving the litter size of Hainan black goats by gene marker-assisted selection.
