摘  要:本试验旨在利用Toll样受体2(TLR2)、Toll样受体4(TLR4)抗体和核因子κB(NF-κB)阻断剂PDTC与刺五加多糖(ASPS)共处理体外培养的淋巴细胞后,观察各种细胞因子mRNA表达量的变化,探究可能的信号通路,从而初步揭示ASPS的免疫调节机制,为将该多糖开发为鸡新型的免疫调节剂提供重要依据。结果显示,与ASPS单独处理组比较,应用TLR2抗体和ASPS共处理后,IL-1βmRNA表达量显著下降(P<0.05),而IL-2、IFN-γ、IL-4、IL-6、IL-10、iNOS和TNF-αmRNA表达量并未显著下降(P>0.05)。TLR4抗体和ASPS共处理后,IL-10、iNOS和TNF-αmRNA表达量显著下降(P<0.05),而IL-1β、IL-2、IFN-γ、IL-4和IL-6 mRNA表达量并未下降,与ASPS单独处理组相近(P>0.05)。另外,应用PDTC和ASPS共处理后,IL-2、IFN-γ和IL-4 mRNA表达量无明显下降,与ASPS单独处理组差异不显著(P>0.05),但是IL-10 mRNA表达量却显著下降(P<0.05)。IL-1β、iNOS和TNF-αmRNA表达量显著下降,并接近空白对照组,IL-6也有所下降,但是与ASPS单独处理组差异不显著(P>0.05)。结果表明ASPS能够通过与TLR2和TLR4结合,活化NF-κB,促进IL-1β、IL-10、iNOS和TNF-α等细胞因子的产生,进而促进Th1细胞的活化、增殖。In attempt to uncover the immunomodulatory mechanism of Acanthopanax senticosus polysaccharide(ASPS),the changes of various cytokine mRNA expression levels in lymphocytes after in vitro co-exposure to TLR2,TLR4 antibody,NF-κB inhibitor PDTC and ASPS were measured.The results showed that after co-treatment with ASPS and TLR2 antibody,IL-1βmRNA expression levels were significantly decreased(P<0.05),while IL-2,IFN-γ,IL-4,IL-6,IL-10,iNOS and TNF-αmRNA expression levels were not significantly decreased(P>0.05).On the other hand,after co-treatment with ASPS and TLR4 antibody,the expression levels of IL-10,iNOS and TNF-αmRNA were significantly decreased(P<0.05),while the expression levels of IL-1β,IL-2,IFN-γ,IL-4 and IL-6 mRNA were not decreased,similar to those in the ASPS group alone(P>0.05).Further,after con-treatment with ASPS and PDTC,the expression levels of IL-2,IFN-γand IL-4 mRNA did not decrease significantly(P>0.05),but the expression levels of IL-10 mRNA decreased significantly(P<0.05).Additionally,IL-1β,iNOS and TNF-αmRNA levels decreased significantly,similar to the blank control group;while IL-6 increased insignificantly(P>0.05).Not much different from the ASPS group.This study indicated that ASPS could bind to TLR2 and TLR4,then activate NF-κB,and promote the production of IL-1β,IL-10,iNOS and TNF-α,thereby promoting Th1 cell activation and proliferation.
