摘 要:试验旨在以群体感应相关性状紫色色杆菌ATCC12472产生的紫色素为评价指标研究红花籽粕对群体感应抑制作用。采用倍比稀释法测定不同终浓度红花籽粕提取物对紫色色杆菌ATCC12472的最小抑菌浓度(MIC)及生长抑制作用;采用指示菌平板法和色素产量抑制法定性、定量检测在亚抑菌浓度下红花籽粕提取物对紫色色杆菌ATCC12472紫色素产生的抑制作用。结果显示,红花籽粕对紫色色杆菌ATCC12472的MIC为300 g/L,亚抑菌浓度下红花籽粕提取物对紫色色杆菌ATCC12472的生长无显著抑制作用(P>0.05);亚抑菌浓度下红花籽粕提取物对紫色色杆菌ATCC12472紫色素的产生均具有抑制作用,其终浓度为25、50、100、200 g/L红花籽粕提取物对紫色色杆菌ATCC12472紫色素的抑制率分别为7.5%、26.4%、32.5%、62.7%。研究表明,红花籽粕中的某些活性物质对紫色色杆菌ATCC12472的群体感应具有抑制作用。The experiment was to study the inhibitory effect of safflower seed meal on quorum sensing by taking the violacein produced by quorum sensing related Chromobacterium violaceum ATCC12472 as the evaluation index.The minimum inhibitory concentration(MIC)and growth inhibition of safflower seed meal extract with different final concentrations on Chromobacterium violaceum ATCC12472 were determined by multiple dilution method.The inhibitory effect of safflower seed meal extract on violacein production of Chromobacterium violaceum ATCC12472 at sub inhibitory concentration was quantitatively detected by indicator plate method and violacein production inhibition method.The results showed that the MIC of safflower seed meal was 300 g/L,and the extract of safflower seed meal had no significant inhibitory effect on the growth of purple Chromobacterium violaceum ATCC12472 at sub inhibitory concentration(P>0.05).Safflower seed meal extract inhibited the production of violacein of Chromobacterium violaceum ATCC12472 at sub inhibitory concentration.The final concentrations were 25,50,100 and 200 g/L,and the inhibition rates of safflower seed meal extract on violacein of Chromobacterium violaceum ATCC12472 were 7.5%,26.4%,32.5% and 62.7%,respectively.The experiment indicates that some active substances in safflower seed meal can inhibit the quorum sensing of Chromobacterium violaceum ATCC12472.
