摘 要:目的:探讨葱白提取物改善非酒精性脂肪性肝病(NAFLD)大鼠糖脂代谢的机制。方法:采用高脂饮食造模,将48只雄性大鼠随机分为空白组、模型组、葱白组、空转组、转染组、转染加葱白组,每组8只,并运用RNAi技术抑制过氧化物酶体增殖物激活受体γ辅激活子1α(PCG-1α)的表达,检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量,HE染色观察肝脏组织病理改变,Western Blot检测肝组织PGC-1α、肉毒碱棕榈酰转移酶1(CPT-1)、中链酰基辅酶A脱氢酶(MCAD)、葡萄糖-6-磷酸酶(G6Pase)、磷酸烯醇式丙酮酸羧激酶(PEPCK)蛋白的表达。结果:与模型组比较,葱白组TC、TG、LDL-C含量显著降低(P<0.05),缓解肝脏脂肪性变,并上调PGC-1α、CPT-1、MCAD、G6Pase、PEPCK蛋白相对表达量(P<0.05)。结论:葱白提取物可增加NAFLD模型大鼠线粒体代谢活性,促进线粒体糖异生及脂肪酸氧化反应,从而起到改善NAFLD模型大鼠糖脂代谢的作用。Objective:To investigate the mechanism by which the fistular onion stalk extract improves glucolipid metabolism in rats with nonalcoholic fatty liver disease(NAFLD).Methods:A high-fat diet modeling was used,48 male rats were randomly divided into normal group,model group,fistular onion group,transfection group,blank transfection group,fistular onion transfection group,8 rats in each group,and the expression of peroxisomal proliferator-activated receptorγcoactivator 1α(PGC-1α)was inhibited by RNAi technology,and serum total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C)were measured.HE staining to observe the histopathological changes in the liver,and the histocyte expression of PGC-1α,carnitine palmitoyltransferase-1(CPT-1),medium-chain acyl-CoA dehydrogenase(MCAD),glucose-6-phosphate kinase(G6 Pase)and phosphoenolpyruvate carboxykinase(PEPCK)proteins were examined by Western Blot.Results:Compared with the model group,fistular onion group could reduce levels of serum TC,TG,LDL-C(P<0.05),relieve liver steatosis,and up-regulated the protein expression of PGC-1α,CPT-1,MCAD,G6 Pase,PEPCK(P<0.05).Conclusion:Fistular onion group could improve glucolipid metabolism in NAFLD rats by increasing mitochondrial metabolic activity,and accelerating gluconeogenesis and fatty acid oxidation.
