摘 要:目的:观察化瘀祛痰方对动脉粥样硬化(AS)家兔心肌组织细胞焦亡的影响,探讨化瘀祛痰方对AS家兔心肌改变的作用及其机制。方法:健康SPF级雄性新西兰白兔45只,随机分为正常组、模型组、化瘀祛痰方组,每组15只。正常组喂饲普通饲料,其余两组喂饲高脂饲料,自由摄食,饮水不限。8周后,化瘀祛痰方组灌胃给予化瘀祛痰方16g·kg^(-1)·d^(-1),正常组与模型组灌胃给予同体积的蒸馏水8mL·kg^(-1)·d^(-1),连续给药4周。HE及Masson染色观察各组家兔心肌组织形态变化,ELISA法检测各组家兔血清及心肌IL-18和IL-1β含量,Western blot法检测各组家兔心肌细胞焦亡相关蛋白表达。结果:与正常组比较,模型组心肌细胞肿胀变性,水肿明显,细胞质内有大量的空泡形成,心肌间质纤维结构紊乱;心肌及血清IL-18和IL-1β含量均显著升高(P<0.01),AIM2、CASP1、NALP3及GSDMD蛋白表达均显著升高(P<0.01);与模型组比较,化瘀祛痰方可以改善心肌形态学变化,显著降低心肌及血清IL-18、IL-1β含量(P<0.01,P<0.05),CASP1、GSDMD及NALP3蛋白表达均显著下降(P<0.01)。结论:化瘀祛痰方可以改善AS家兔心肌组织的变化,其机制可能与调节细胞焦亡存在密切关系。Objective:Observe the effects of Huayu Qutan Decoction(HYQT)on pyroptosis of cardiomyocyte in atherosclerotic(AS)rabbits,and to explore the effect and mechanism of HYQT on cardiomyocyte changes in AS rabbits.Methods:A total of 45 SPF male New Zealand white rabbits were randomly divided into control group,model group and HYQT group with 15 rabbits in each group.The normal group was fed with normal diet,and the other groups were fed with high-fat diet.After 8 weeks,the HYQT group was given corresponding drug intervention,and the dosage was converted according to the conversion coefficient of human and animal body surface area.The dosage of HYQT group was 16 g·kg^(-1)·d^(-1),while the normal group and model group were given the same volume of distilled water by gavage,with the dosage volume of 8 mL·kg^(-1)·d^(-1) for 4 weeks.He and Masson staining were used to observe the morphological changes of myocardial tissue.ELISA method was used to detect the contents of IL-18 and IL-1βin serum and myocardium.The expression of pyroptosis protein in cardiomyocyte was detected by Western blot.Results:Compared with control group,myocardial cells in model group were swollen and degenerated,edema was obvious,a large number of vacuoles were formed in cytoplasm,myocardial interstitial fiber structure was disordered;the contents of IL-18 and IL-1βin myocardium and serum were increased(P<0.01),and the protein expressions of AIM2,caspase1,NALP3 and GSDMD were increased(P<0.01);compared with model group,HYQT group could improve the above myocardial morphological changes,the contents of IL-18 and IL-1βin myocardium and serum were decreased(P<0.01,P<0.05),the protein expression of CASP1,GSDMD and NALP3 decreased(P<0.01).Conclusion:Huayu Qutan Decoction can improve the changes of myocardial tissue in as rabbits,which may be closely related to the regulation of pyroptosis.
