摘 要:目的:研究人参白术枣仁颗粒健脾养心、宁心安神和改善睡眠功能的药理作用及其作用机制。方法:采用负重游泳法复合药物诱导建立心脾两虚证失眠小鼠模型,观察人参白术枣仁颗粒对其体质量、小肠推进、悬尾不动时间、游泳时间、睡眠行为,以及脑中神经递质含量的影响。观察人参白术枣仁颗粒对正常小鼠自主活动和睡眠功能的影响。结果:与模型组比较,中药高剂量组能改善模型小鼠心脾两虚症状,延长游泳时间和睡眠时间、减少悬尾不动时间,增加5-HT含量、降低Glu含量(P<0.05,P<0.01),能减少正常小鼠自主活动次数、延长睡眠时间、缩短睡眠潜伏期(P<0.05,P<0.01)。结论:人参白术枣仁颗粒有健脾养心、宁心安神和改善睡眠功能作用,其作用机制可能与调节脑内5-HT、Glu含量有关。Objective:To study the pharmacological effects and mechanism of Renshen Baizhu Zaoren Granules on invigorating spleen and nourishing heart,quieting the heart and tranquilizing mind,improving sleep function.Methods:The mice model of heart-spleen deficiency syndrome was established by weight-bearing swimming method combined with drugs induction,and the effects of Renshen Baizhu Zaoren Granules on body weight,intestinal propulsion,tail suspension immobility,swimming time,sleep behavior,and the content of neurotransmitters in the brain were observed.Effects of Renshen Baizhu Zaoren Granules on autonomous activity and sleep function of normal mice were observed.Results:Compared with model group,Chinese medicine high-dose group could improve the symptoms of deficiency of heart-spleen in model mice,prolong swimming time and sleep time,decrease tail immobile time,increase 5-HT content and decrease Glu content(P<0.05,P<0.01).In addition,it could reduce the number of autonomous activities,prolong the sleep time and shorten the sleep latency of normal mice(P<0.05,P<0.01).Conclusion:Renshen Baizhu Zaoren Granules has the effects of invigorating spleen and nourishing heart,quieting the heart and tranquilizing mind,improving sleep function,and its mechanism may be related to the regulation of the contents of 5-HT and Glu in brain.
