摘  要:目的:观察参茸丸对卵巢卵泡生长发育的影响,对下丘脑-垂体-卵巢轴激素调节的作用以及对卵巢性激素受体的影响,以期探讨参茸丸对排卵障碍型不孕症的治疗作用。方法:采用丙酸睾酮所致排卵障碍型不孕大鼠模型,造模成功大鼠按体质量随机分为模型组,枸橼酸氯米芬组,参茸丸高、中、低剂量组,大鼠连续给药6周。采用HE染色及免疫组化染色检测大鼠卵巢、子宫病理形态及卵巢雌激素受体α(ERα)、促卵泡激素受体(FSHR)、促黄体生成素受体(LHR)蛋白表达情况;采用放射免疫分析法检测大鼠血清雌二醇(E_(2))、促黄体生成素(LH)、卵泡生成素(FSH)、雄激素(T)、泌乳素(PRL)、孕激素(P)及及促性腺激素释放激素(GnRH)激素水平;采用实时荧光定量PCR方法检测卵巢组织ERα、FSHR、LHR mRNA表达水平。结果:与模型组比较,参茸丸各组大鼠卵巢初级卵泡数和次级卵泡数均有增加(P<0.05,P<0.01),参茸丸中剂量组大鼠子宫内膜厚度显著增加(P<0.05),血清T、E_(2)、LH、FSH及PRL激素水平显著升高(P<0.05,P<0.01),血清GnRH激素水平显著降低(P<0.01),卵巢组织FSHR蛋白及ERαmRNA表达量显著增加(P<0.05)。结论:参茸丸可以增强排卵障碍型不孕大鼠性腺轴激素调节作用,改善排卵障碍型不孕大鼠卵巢功能和子宫受孕环境。Objective:To observe the effects of Shenrong Pills on the growth and development of ovarian follicles,the regulation of gonadal axis hormones and the effects on ovarian sex hormone receptors,in order to explore the therapeutic effects of Shenrong Pills on infertility.Methods:Using testosterone-induced ovulation dysfunction infertility rat model,the successfully modeled rats were randomly divided into model group,Clomiphene Citrate group,Shenrong Pills high,medium,and low dose groups according to their body weight.The rats were continuous administration for 6 weeks.HE staining and immunohistochemical staining were used to detect the pathological morphology of ovary and uterus and the expression of ERα,FSHR,LHR protein;radioimmunoassay was used to detect rat serum E_(2),LH,FSH,T,PRL,P and GnRH hormone level;real-time fluorescent quantitative RT-PCR method was used to detect the expression of ERα,FSHR and LHR mRNA in ovarian tissue.Results:Compared with the model group,the number of primary ovarian follicles and secondary follicles in each group of Shenrong Pills increased(P<0.05,P<0.01),and the endometrial thickness of rats in the middle dose group of Shenrong Pills increased(P<0.05),the serum T,E_(2),LH,FSH and PRL hormone levels in the middle does group of Shenrong Pills increased(P<0.05,P<0.01),the serum GnRH hormone level decreased(P<0.01),and the expression of FSHR protein and ERαmRNA in ovarian tissue increased(P<0.05).Conclusion:Shenrong Pills can enhance the hormone regulation of the gonadal axis in ovulatory infertile rats,and improve the ovarian function and uterine conception environment in ovulatory infertile rats.
