摘 要:本试验应用液相色谱-质谱联用(LC-MS)技术分析乳鸽发育期间血清代谢物的变化。选取14日龄、体重(398.33±43.28)g与21日龄、体重(449.17±49.44)g的健康乳鸽各6只,翅静脉采血,分离血清作为样本,采用基于LC-MS非靶向代谢组学技术进行分析,根据变量投影重要度(VIP)和独立样本t检验筛选出差异代谢物,并作通路富集分析。结果显示:在2个日龄乳鸽血清样本中共检测到11538个特征代谢峰,正、负离子模式下共鉴定出694种代谢物。经搜库(自建库、Metlin、HMDB等数据库)定性分析,共筛选出71种差异代谢物,其中有49种差异代谢物丰度显著上调(P<0.05),22种差异代谢物丰度显著下调(P<0.05)。KEGG通路富集共获得29条代谢通路,其中14条代谢通路差异显著(P<0.05),主要为甘油脂代谢、醚脂代谢、糖基磷脂酰肌醇(GPI)-锚定生物合成等。乳鸽血清S-腺苷蛋氨酸、L-棕榈酰肉碱、硬脂酰肉碱和磷脂类代谢物的含量随着日龄增加而显著增加(P<0.05)。上述结果有助于了解乳鸽发育期生理状态的变化,为生产中更好地满足其营养需要提供参考。The changes of serum metabolites during the development of squabs were analyzed with the technique of liquid chromatography-mass spectrometry.Six 14-day-old healthy squabs with the body weight of(398.33±43.28)g and six 21-day-old healthy squabs with the body weight of(449.17±49.44)g were selected for wing venous blood sampling,and serum was prepared and analyzed with non-target metabolomics based on the technique of LC-MS.The differential metabolites were screened out according to the variable importance in projection(VIP)and independent sample t-test,and their pathway enrichments were analyzed.The results showed that a total of 11538 characteristic metabolic peaks were detected in the serum of 14-and 21-day-old squabs,and 694 metabolites identified in the positive and negative ion models.After the qualitative analysis of database(self-built database,Metlin,HMDB etc.),a total of 71 differential metabolites were screened,of which 49 differential metabolites were significantly up-regulated(P<0.05),and 22 differential metabolites were significantly down-regulated(P<0.05).The KEGG pathway enrichment resulted in 29 metabolic pathways,14 of which were significantly different(P<0.05),mainly glycerophospholipid metabolism,ether lipid metabolism,glycosylphosphatidylinositol(GPI)-anchor biosynthesis etc.The contents of serum S-adenosylmethionine,L-palmitoylcarnitine,stearoylcarnitine and some metabolites of phospholipids were significantly rosed with the increase of age of squabs(P<0.05).The above data are helpful to understand the changes of the physiological state of the squabs during the development and provide references for better satisfying their nutritional needs in production.
