摘  要:用纯化的塞尼卡病毒(Seneca Valley virus,SVV)VP1蛋白,辅以不同佐剂免疫小鼠,分别从体液免疫及细胞免疫对小鼠的免疫效果进行评估。结果表明,VP1-AL组与VP1-FCA组的特异性抗体一免后7 d均呈显著上升趋势,PBS组变化不明显。二免后14 d,VP1-FCA组特异性抗体效价达到最高值(1∶163840),PBS组抗体效价是1∶16(P<0.01);VP1-AL组特异性抗体效价达到最高值(1∶5120),PBS组抗体效价是1∶16(P<0.01)。中和抗体效价,二免后7 d VP1-FCA组达到最高值(1∶512),PBS组抗体效价是1∶8(P<0.01);二免后14 d VP1-AL组达到最高值(1∶1024),PBS组抗体效价是1∶16(P<0.01)。小鼠T淋巴细胞亚群CD3^(+)CD4^(+)与CD3^(+)CD8^(+)在二免后VP1-FCA组显著升高(P<0.05),VP1-AL组极显著升高(P<0.01)。淋巴细胞增殖试验表明各试验组均显著升高(P<0.05)。对免疫小鼠血清中细胞因子检测结果表明,IL-4的分泌显著提高(P<0.05),IL-2与IFN-γ变化不明显。本研究利用SVV VP1重组蛋白联合佐剂,VP1-AL组是有前景的候选疫苗,可诱导小鼠体内产生免疫应答且效果良好,为SVV疫苗猪体免疫研究奠定理论基础。Purified Seneca Valley virus(SVV)VP1 protein was used to immunize mice with different adjuvants,the immune effects of mice were evaluated by humoral immunity and cellular immunity,respectively.The results showed that the specific antibody levels of VP1-AL group and VP1-FCA group increased significantly 7 d after the first immunization.The titer of specific antibodies of VP1-FCA group reached the highest(1∶163840)at 14 d after the second immunization,and that of the PBS group was 1∶16(P<0.01).The specific antibodies titer of the VP1-AL group reached the highest value at 14 d after the second immunization(1∶5120),and that of the PBS group was 1∶16(P<0.01).The titer of neutralizing antibody was significantly higher than that of PBS group at 7 d after the second immunization,VP1-FCA group reached its maximum(1∶512),PBS antibody titers were 1∶8.The highest value was reached in VP1-AL group at 14 d after the second immunization and that of the PBS group was 1∶16(P<0.01).T lymphocyte subsets CD3^(+)CD4^(+)and CD3^(+)CD8^(+)were significantly increased in VP1-FCA group and extremely significantly increased in VP1-AL group after second immunization(P<0.01).Lymphocyte proliferation assay showed that the increase was significant in all immunized groups(P<0.05).The serum cytokines in immunized mice showed that the secretion of IL-4 was significantly increased(P<0.05),while the changes of IL-2 and IFN-γwere not significant.In this study,SVV VP1 recombinant protein combined with adjuvant was used,and the VP1-AL group was a promising candidate vaccine,which could induce immune response in mice with good effect,laying a foundation for the study of SVV vaccine in swine immunization.
