饲料中添加沸石对慢性肾衰犬结肠尿素转运蛋白B及氨转运体蛋白Rh表达的影响

摘　　要：本研究旨在探究沸石对慢性肾衰犬结肠尿素转运蛋白B及氨转运体蛋白Rh表达的影响。选取15条2岁左右的中华田园犬,采用肾动脉结扎制作慢性肾衰(CRF)模型,另取5只犬仅腹壁切开作为假手术对照组(SOG)。造模成功后,随机等分成3组:模型组(MD)、低剂量沸石组(LCD)和高剂量沸石组(HCD),SOG组与MD组饲喂基础饲料,LCD组为1%沸石饲料,HCD为5%沸石饲料,试验连续4周。用自动化血液生化仪检测血清尿素氮(BUN)和粪尿素氮(FUN),用谷氨酸脱氢酶法测定门静脉血氨,用尿素检测试剂盒测定尿尿素量(uUrea),qRT-PCR检测结肠黏膜尿素转运蛋白B(UT-B)、氨转运体蛋白b(Rhbg)与氨转运体蛋白c(Rhcg)mRNA表达,Western blot法检测UT-B、Rhbg与Rhcg的蛋白表达。结果显示,MD组的BUN、FUN及门静脉血氨均高于SOG组,uUrea降低(P<0.01),与MD组比较,HCD组的BUN、FUN、门静脉血氨均显著降低(P<0.05);MD组Rhbg、Rhcg的mRNA表达显著高于SOG组(P<0.05),与MD组比较,HCD组Rhbg、Rhcg及LCD组Rhcg的mRNA表达显著下调(P<0.05),各组间UT-B的mRNA表达无显著变化(P>0.05);MD组Rhbg、Rhcg的蛋白表达显著高于SOG组(P<0.05),与MD组比较,HCD组Rhbg、Rhcg的表达降低,MCD组Rhcg表达降低,差异显著(P<0.05),各组间UT-B表达无差异;Pearson相关性分析发现,慢性肾衰犬BUN水平与Rhcg呈强的正相关(r=0.855,P<0.05),与Rhbg呈中等正相关(r=0.769,P<0.05)。以上结果表明,沸石具有促进CRF犬BUN经肠排泄的作用,与结肠黏膜Rhbg、Rhcg的表达下调有关,以添加5%剂量最佳。In order to studythe effect of zeolite added to diet on the expression of urea transporter B and ammonia transporter Rh in colon of dogs with chronic renal failure,a total of 152-year-old Chinese garden dogs were used to established chronic renal failure model by ligation of renal artery,and another 52-year-old Chinese garden dogs were only opened abdominal wall as the sham operation control group(SOG)fed with basic diet.After successful modeling,all dogs with CRF were randomly divided into 3 groups:model group(MD)fed with basic diet,low zeolite dose group(LCD)fed with 1%zeolite diet,and high zeolite dose group(HCD)fed with 5% zeolite diet.Test lasted 4 weeks.Serum urea nitrogen(BUN)and fecal urea nitrogen(FUN)were measured by automatic blood biochemical instrument.Ammonia in portal vein were measured by glutamate dehydrogenase.Urea in urine were measured by urea test kit.Expression of mRNA of urea transporter B(UT-B),ammonia transporter b(Rhbg)and ammonia transporter c(Rhcg)in colonic mucosa were detected by qRT-PCR.Protein expression of UT-B,Rhbg and Rhcg were detected by Western blot.The results showed:BUN,FUN and blood ammonia of portal vein in the MD group were higher than those in the SOG group,and urea in urine was lower than that in the SOG group(P<0.01).Compared with the MD group,the levels of BUN,FUN and ammonia of portal vein in the HCD group decreased significantly(P<0.05).The mRNA expressions of Rhbg and Rhcg in MD group were significantly higher than those in the SOG group(P<0.05).Compared with MD group,The mRNA expressions of Rhbg and Rhcg in the HCD group decreased significantly(P<0.05),while that of Rhcg in LCD group decreased significantly(P<0.05).There was no significant change in the mRNA expression of UT-B among all groups(P>0.05).The expressions of Rhbg and Rhcg in the MD group was significantly higher than those in the SOG group(P<0.05).Compared with MD group,the expressions of Rhbg and Rhcg in the HCD group and that of Rhcg in the LCD group significantly decreased(P<0.05).There was no diff
