摘  要:目的分析男性和女性系统性红斑狼疮(SLE)患者中血浆的差异表达蛋白及其生物学功能。方法纳入SLE男性、SLE女性、健康男性、健康女性各8例,收集其外周血标本并分离血浆。利用同位素标记相对和绝对定量(iTRAQ)技术筛选SLE男性与SLE女性之间、SLE女性与健康女性之间、SLE男性与健康男性之间的差异表达蛋白谱,并对所有的差异表达蛋白进行基因本体论(GO)分析和京都基因与基因组百科全书(KEGG)分析。结果 (1)SLE女性与SLE男性之间的差异表达蛋白共277个(表达上调193个,表达下调84个),涉及的生物过程主要包括生物调节、免疫系统过程、生物过程调节等,涉及的信号通路包括类风湿关节炎、原发性免疫缺陷及生产IgA的肠道免疫网络等。(2)SLE女性与健康女性之间的差异表达蛋白共242个(表达上调181个,表达下调61个),涉及的生物过程主要包括细胞过程、免疫系统过程、生物过程调节等,涉及的信号通路包括类风湿关节炎、FcεRI信号通路及麻疹等。(3)SLE男性与健康男性之间的差异表达蛋白共214个(表达上调114个,表达下调100个),涉及的生物过程主要包括免疫系统过程、刺激反应、细胞过程等,涉及的信号通路主要包括心肌收缩、钙离子信号通路及过氧化物酶体增殖物激活受体信号通路等。结论 SLE患者的血浆蛋白表达存在性别差异,并且差异表达蛋白所参与的生物学过程及涉及的信号通路也存在差异。Objective To analyze plasma differentially expressed proteins and their biological function in male and female patients with systemic lupus erythematosus(SLE).Methods Eight SLE men,eight SLE women,eight healthy men,and eight healthy women were enrolled,their peripheral blood samples were collected and plasma separation was conducted.Isobaric tags for relative and absolute quantitation(iTRAQ) technique was used to screen the profiles of differentially expressed proteins between SLE men and SLE women,between SLE women and healthy women,and between SLE men and healthy men.In addition,Gene Ontology(GO) analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) analysis were performed on all of the differentially expressed proteins.Results(1) Totally 277 differentially expressed proteins were seen between SLE women and SLE men(193 up-regulated and 84 down-regulated),the biological processes involved mainly included biological modulation,immune system process,and modulation of biological process,the signaling pathways involved included rheumatoid arthritis,primary immunodeficiency and IgA-producing gut immunity network.(2) There were 242 differentially expressed proteins between SLE women and healthy women(181 up-regulated and 61 down-regulated),the biological processes involved mainly included cellular process,immune system process and modulation of biological process,and the signaling pathways involved included rheumatoid arthritis,FcεRI signaling pathway and measles.(3) A total of 214 differentially expressed proteins were observed between SLE men and healthy men(114 up-regulated and 100 down-regulated),the biological processes involved mainly included immune system process,stimulus response and cellular process,the signaling pathways involved commonly included myocardial contraction,calcium ion signaling pathway and peroxisome proliferator-activated receptor signaling pathway.Conclusion Among SLE patients,the expression of plasma proteins exhibits a gender-specific difference,and differentially expressed proteins
