摘 要:目的探讨鱼腥草总黄酮对骨质疏松症大鼠过氧化和免疫失调的作用机制。方法将75只雌性SD大鼠均分为假手术组、模型组、鱼腥草总黄酮低、中、高剂量组,除假手术组外其余各组大鼠均采用双侧卵巢切除术制备骨质疏松症大鼠模型,鱼腥草总黄酮低、中、高剂量组术后分别给予0.5、1.0、2.0 g/kg鱼腥总黄酮灌胃干预,假手术组和模型组大鼠灌胃等体积生理盐水,连续干预4周。比较各组大鼠骨体积分数(BV/TV)、特定骨表面积(BSA/BV)、骨小梁厚度(Tb.Th)、骨小梁分离度(Tb.Sp)、骨小梁和软骨含量,检测骨组织中破骨细胞相关受体(OSCAR)、抗酒石酸酸性磷酸酶(TRAP)基因和蛋白表达水平,ELISA法检测骨组织超氧化物歧化酶(SOD)、丙二醛(MDA)、还原型谷胱甘肽(GSH)水平以及外周血白介素-17(IL-17)、白介素-23(IL-23)、白介素-10(IL-10)含量。结果与假手术组相比,模型组BV/TV、Tb.Th、骨小梁和软骨含量、骨组织SOD水平降低(P<0.05),BSA/BV、Tb.Sp、OSCAR和TRAP基因及蛋白水平升高(P<0.05),骨组织MDA、GSH水平升高(P<0.05),骨组织和外周血IL-17、IL-23、IL-10含量升高(P<0.05);与模型组相比,鱼腥草总黄酮中、高剂量组BV/TV、Tb.Th、骨小梁和软骨含量、骨组织SOD水平升高(P<0.05),BSA/BV、Tb.Sp、OSCAR和TRAP基因及蛋白水平降低(P<0.05),骨组织MDA、GSH水平降低(P<0.05),骨组织和外周血IL-17、IL-23水平降低(P<0.05),骨组织和外周血IL-10含量升高(P<0.05)。结论鱼腥草总黄酮可以降低骨质疏松症大鼠过氧化和免疫反应,改善成/破骨细胞间的功能平衡。Objective To explore the possible action mechanism of total flavonoids from houttuyniacordata(TFHC) on protecting from peroxidation and immune disorders in rats with ovariectomy induced osteoporosis.Methods A total of 75 female SD rats were divided into sham operation group, model group, low-dose, middledose and high-dose TF-HC groups. Except sham operation group, bilateral oophorectomy was conducted in the other groups to generate osteoporosis rat models. After surgery, low-dose, middle-dose and high-dose TF-HC groups were given intragastric administration of 0.5, 1.0 and 2.0 g/kg total flavonoids from houttuyniacordata,while sham operation group and model group were given the same volume of normal saline for 4 weeks. The bone volume/total volume(BV/TV), trabecular bone surface/bone volume(BSA/BV), trabecular bone thickness(Tb.Th), trabecular bone space(Tb.Sp), and contents of trabecular bones and cartilages were compared among all groups. The expression levels of osteoclast-associated receptor(OSCAR) and tartrateresistant acid phosphatase(TRAP) genes and proteins in bone tissues were detected. The levels of superoxide dismutase(SOD),malondialdehyde(MDA) and reduced glutathione(GSH) in bone tissues, and peripheral blood interleukin-17(IL-17), interleukin-23(IL-23) and interleukin-10(IL-10) were detected by ELISA. Results Compared with sham operation group, BV/TV, Tb. Th, contents of trabecular bones and cartilages, and SOD level were decreased in model group(P<0.05). BSA/BV, Tb.Sp, expression levels of OSCAR and TRAP genes and proteins were increased(P<0.05), levels of MDA and GSH in bone tissues were increased(P<0.05), and contents of IL-17, IL-23 and IL-10 in bone tissues and peripheral blood were increased in mice of model group(P<0.05). Compared with model group, BV/TV, Tb.Th, contents of trabecular bones and cartilages, and SOD level were increased in middle-dose and high-dose TF-HC groups(P<0.05). BSA/BV, Tb.
